单增李斯特菌致流产小鼠子宫中Th1和Th2细胞因子的变化

【摘要】 为探讨Th1和Th2细胞因子在单增李斯特菌(Listeria monocytogenes,LM)致小鼠流产中的作用,用LM感染妊娠小鼠和小鼠巨噬细胞,统计胚胎存活率,酶联免疫测定(enzyme linked lmmuno sorbent assay,ELISA)小鼠子宫组织中和巨噬细胞培养基中主要的Th1和Th2细胞因子水平.结果显示,LM诱导的流产小鼠子宫中Th1细胞因子水平显著增加,溶血素O (listeriolysin O,LLO)基因缺陷型(Δhly)LM感染组胚胎存活率和子宫中Th1、Th2细胞因子水平无显著变化;LM感染的巨噬细胞分泌的Th1细胞因子和白细胞介素-4(interleukin, IL-4)水平显著增加,IL-10水平显著降低,Δhly LM感染组Th1和Th2细胞因子水平无显著变化.因此,LM感染后子宫中Th1/Th2细胞因子失衡诱导了小鼠流产,这可为临床防治LM诱导的流产提供科学依据.

【Abstract】 To explore whether Th1 and Th2 cytokines play important roles in Listeria monocytogenes(LM)-induced mouse abortion,the pregnant mice and mouse macrophages were infected with LM. Embryo survival rates were calculated. Th1 and Th2 cytokine levels were analyzed in uterus and medium of mouse macrophages. Results showed that Th1 cytokine levels increased significantly in uterus of abortive mice induced by LM. Embryo survival rates and Th1 and Th2 cytokine levels showed no changes in uterus of mice infected by listeriolysin O(LLO)-deficient LM(Δ hly LM). Th1 cytokine and interleukin(IL)-4 levels increased while IL-10 level decreased in the medium of macrophages infected by wild-type LM.After infected byΔhly LM,Th1 and Th2 cytokine levels showed no changes.Therefore,LM-induced Th1/Th2 imbalance in uterus can causes the mouse abortion,which provides scientific basis for clinical prevention and treatment of LM-induced abortion.
