MMP-2、MMP-9在系统性硬化病小鼠模型中的表达及意义

【摘要】 目的探讨基质金属蛋白酶2 (MMP-2)和基质金属蛋白酶9 (MMP-9)在系统性硬化病(SSc)小鼠模型中的表达。方法选取16只雌性BALB/c小鼠,随机分成对照组与SSc组,每组8只。SSc组小鼠给予博莱霉素,对照组给予磷酸缓冲盐水(PBS)处理。采用HE、Masson三色染色法观察小鼠皮肤和肺部病理改变,测定羟脯氨酸(HYP)的含量。荧光定量PCR法检测皮肤、肺组织中MMP-2和MMP-9的mRNA含量。结果与SSc组相比,模型组纤维化和炎症反应加重(P<0.05),HYP含量增加(P<0.05),MMP-2和MMP-9的mRNA表达量降低(P<0.05)。结论 MMP-2、MMP-9在博来霉素诱导的SSc小鼠模型皮肤、肺组织中表达量增高,MMP-2、MMP-9可能在SSc发生发展中起重要作用。

【Abstract】 Objective To investigate the expression of matrix metalloproteinase 2(MMP-2) and matrix metalloproteinase 9(MMP-9) in a mouse model of systemic sclerosis(SSc).Methods Sixteen female BALB/c mice were randomly divided into two groups which were the control group and the SSc group and with eight in each group.Mice in the SSc group were treated with bleomycin(BLM),and the control group was treated with phosphate buffered saline(PBS).HE and Masson trichrome staining were used to observe the skin and lung pathological changes in mice,and the content of hydroxyproline(HYP) was determined.The mRNA content of MMP-2 and MMP-9 in skin and lung tissue was detected by real-time PCR.Results Compared with the SSc group,fibrosis and inflammatory response worsened in the model group(P<0.05),HYP content increased(P <0.05),and mRNA expression of MMP-2 and MMP-9 decreased(P<0.05).Conclusion MMP-2 and MMP-9 are increased in the skin and lung tissues of SSc mouse models induced by bleomycin,and MMP-2 and MMP-9 may play an important role in the development of SSc.
