循环肿瘤DNA在肝细胞癌早期诊断中的研究进展

【摘要】 监测肝细胞的恶性转化对肝细胞癌的早期诊断和及时干预至关重要。肿瘤细胞主动或被动释放核酸片段进入血液循环中,成为循环肿瘤DNA(circulating tumor DNA,ctDNA),其携带有与肿瘤细胞一致的遗传信息。近几年,随着新一代基因检测技术的发展,基于"液体活检术"的ctDNA检测以其独特的优势逐渐成为研究热点,被认为是新一代肿瘤分子标志物,现从循环细胞游离DNA(circulating cell-free DNA,cfDNA)浓度、ctDNA基因突变和ctDNA的甲基化改变来阐述ctDNA检测在肝细胞癌早期诊断中的作用。

【Abstract】 Monitoring the malignant transformation of hepatocytes is very important for the early diagnosis and timely intervention of hepatocellular carcinoma. Tumor cells actively or passively release nucleic acid fragments into the blood circulation, becoming circulating tumor DNA(ctDNA), which carries genetic information consistent with tumor cells. In recent years, with the development of next-generation gene detection technology, ctDNA detection based on "liquid biopsy" has gradually become a research hotspot with its unique advantages, considered as a new generation of tumor molecular marker. In this review, the role of ctDNA detection in the early diagnosis of hepatocellular carcinoma is discussed from the concentration of circulating cell free DNA(cfDNA), mutation of ctDNA gene and methylation of ctDNA.
