【摘要】 目的探讨大肠癌(含黏液腺癌及非黏液腺癌)在癌变过程中Fascin和Galectin-3的表达及临床意义。方法收集江门市中心医院2014年1月~2015年12月经手术切除的大肠癌患者的临床病理资料,采用免疫组织化学(S-P)法检测76例大肠癌、34例大肠腺瘤及34例正常大肠黏膜组织中Fascin与Galectin-3蛋白水平变化,分析Fascin及Galectin-3蛋白的表达与大肠癌临床病理因素的关系,并对两者进行相关性分析。结果 Fascin在大肠癌、大肠腺瘤及正常大肠黏膜组织中的阳性表达率分别为47.37%、11.76%和2.94%,三组间差异有统计学意义(P <0.01);Galectin-3在上述三组组织中的阳性表达率分别为68.42%、38.24%和17.65%,差异有统计学意义(P <0.01);Fascin与Galectin-3在大肠癌组织中的表达呈正相关,并且两者高表达与TNM分期、淋巴结转移及肿瘤的分化程度有关(P <0.05),与患者的性别、年龄、是否为黏液型腺癌等组织学类型无关。结论 Fascin和Galectin-3参与了大肠癌的侵袭转移,为大肠癌的分子靶向治疗提供了新思路。

【Abstract】 Objective To investigate the expression of Fascin and Galectin-3 in colorectal cancer(CC, including mucinous adenocarcinoma and non-mucinous adenocarcinoma) during carcinogenesis and the clinical significance. Methods Clinicopathological data of the CC patients who underwent surgical resection in Jiangmen Central Hospital from January 2014 to December 2015 were collected. Immunohistochemical(S-P) method was used to detect the changes of Fascin and Galectin-3 protein levels in 76 cases of CC, 34 cases of colorectal adenoma and 34 cases of normal colorectal mucosa tissues. The relationship between the expression of Fascin and Galectin-3 protein and clinicopathological factors of CC was analyzed, and the correlation between the two was analyzed. Results The positive expression rate of Fascin in CC, colorectal adenoma and normal colorectal mucosa tissues was 47.37%, 11.76% and 2.94% respectively, with statistically significant differences among the three groups(P < 0.01). The positive expression rate of Galectin-3 in the above three groups of tissues was 68.42%, 38.24% and 17.65% respectively, with statistically significant differences(P < 0.01). The expression of Fascin and that of Galectin-3 were positively correlated in CC tissues, and the high expression of Fascin and Galectin-3 was related to TNM staging, lymph node metastasis and tumor differentiation(P < 0.05) and had nothing to do with the patients’ sex, age, and histological types, e.g. whether it was mucinous adenocarcinoma or not. Conclusion Fascin and Galectin-3 are involved in the invasion and metastasis of CC, which provides new trains of thought for molecular targeted therapy of CC.
