杨桃根提取物对肝纤维化大鼠肝脏显微结构的影响

【摘要】 目的:探讨杨桃根提取物(EACR)对肝纤维化(HF)大鼠肝脏显微结构的影响。方法:适应性喂养7 d后,将84只SD大鼠随机分为6组。除正常对照组给予橄榄油外,其余各模型组给予50%四氯化碳(CCl4)橄榄油建立HF模型,共12周。第9~12周,各组给予相应药物干预治疗。实验结束后,检测血清中肝功能的标志性指标:白蛋白(ALB)、球蛋白(GLB)、总胆红素(TBIL)和总胆固醇(TC)水平。免疫组化法观察肝脏组织中核转录因子κB(NF-κB)、B淋巴细胞瘤-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)蛋白的表达情况。透射电镜下观察肝脏组织超微结构。结果:与模型组相比,EACR给药组显著上调白蛋白/球蛋白(A/G)的比值,显著下调TBIL、TC的水平。免疫组化结果显示,EACR给药组显著下调NF-κB、Bax的表达量,上调Bcl-2的表达量。从透射电镜结果可知,模型组大鼠肝细胞结构形态明显改变,细胞核溶解消失,细胞质中的细胞器肿胀变形或溶解。EACR给药组肝脏组织超微结构较模型组明显改善。结论:EACR对CCl4诱导的HF大鼠肝脏细胞具有保护作用。

【Abstract】 Objective: To investigate the effect of extract of Averrhoa carambola L. root(EACR) on the liver microstructure of liver fibrosis(HF) rats. Methods: After 7 days of adaptive feeding, 84 SD rats were randomly divided into 6 groups. Except for the normal control group wasgiven olive oil, the remaining model groups were given 50% carbon tetrachloride(CCl4) olive oil to establish the HF model for a total of 12 weeks. From the 9 th to the 12 th week, each group was given corresponding drug intervention treatment. After the experiment, the levels of serum liver function markers including albumin(ALB), globulin(GLB), total bilirubin(TBIL), and total cholesterol(TC) were detected. Immunohistochemical method was used to observe the expression of nuclear factorκB(NF-κB), b-cell lymphoma 2(bcl-2) gene,and bcl-2-associated X protein(Bax) protein in liver tissue. The ultrastructure of liver tissue was observed under transmission electron microscope. Results: Compared with the model group, the albumin/globulin(A/G) ratio was significantly up-regulatedwhile the levels of TBIL and TCweredown-regulated significantlyin the EACR groups. Immunohistochemical results showed that the expression levels of NF-κB and Bax were significantly down-regulatedwhile the expression of bcl-2 was up-regulatedin the EACR groups. From the results of transmission electron microscopy, it could be seen that the structure and shape of the liver cells of the model group obviously changed, the nucleolysiswas observed, and the organelles in the cytoplasm were swollen and deformed or dissolved. The ultrastructure of liver tissue in the EACR groups was significantly improved compared with the model group. Conclusion: EACR has a protective effect on liver cells of rats withHF induced by CCl4.
