SDF-1/CXCR4信号轴在牙髓血运重建中的作用
The role of SDF-1/CXCR4 signal axis in dental pulp revascularization
【摘要】 目的:研究基质细胞衍生因子-1/CXC趋化因子受体4(SDF-1/CXCR4)信号轴在牙髓血运重建中对组织再生的作用。方法:18只8周龄SD大鼠随机分为常规组(N组)、常规+SDF-1水凝胶组(N+SDF-1组)和常规+抗SDF-1抗体中和作用组(N+antiSDF-1组),每组6只,实验牙为双侧下颌第一磨牙,建立无髓根管模型后,按照分组进行处理。14 d后,收集双侧下颌骨,分别进行苏木精-伊红(HE)染色和免疫组织化学检测,观察根管新生组织类型以及CXCR4+细胞情况。结果:N组、N+SDF-1组根管新生组织冠方为不规则矿化基质,根尖方向可见成纤维样细胞组成的致密结缔组织,N+SDF-1组部分根管内可见新生小血管;N+anti SDF-1组典型组织学表现为根管内无明显的新生结缔组织,根尖孔区有大量的幼稚成纤维样细胞和小血管集聚,细胞无进入根管内的趋势。N组和N+SDF-1组根管内CXCR4+细胞,N+anti SDF-1组根管内见少量CXCR4+细胞。结论:SDF-1/CXCR4信号轴在牙髓血运重建中通过趋化细胞归巢进入根管内的方式,参与了组织再生,并具有促进血管新生的作用。
【Abstract】 Objective: To study the effect of matrix cell-derived factor-1/CXC chemokine receptor 4(SDF-1/CXCR4)signal axis on tissue regeneration in dental pulp revascularization. Methods: Eighteen 8-week-old SD rats were randomly divided into three groups: routine group(N group),routine+SDF-1 hydrogel group(N+SDF-1 group),and routine+ anti-SDF-1 antibody neutralization group(N + antiSDF-1 group),with 6 rats in each group. The experimental teeth were bilateral mandibular first molars. After the medulleless root canal model was established,the teeth were treated according to the grouping. In group N,dental pulp revascularization;in N+SDF-1 group,SDF-1 hydrogel was implanted into the root canal on the basis of dental pulp revascularization;in group N+anti SDF-1,anti-SDF-1 antibody was injected locally in the vestibular sulcus and the root canal on the basis of dental pulp revascularization. After 14 days of treatment,bilateral mandibles were collected for HE staining and immunohistochemical detection,and the new tissue types of root canals and CXCR4+ cells were observed. Results: In N group and N+SDF-1 group,the new tissue of root canal was irregular mineralized matrix,the dense connective tissue composed of fibroblast-like cells could be seen in the apical direction;and in N + SDF-1 group,the new small vessels could be seen in some root canals;while in N + anti SDF-1 group,there was no obvious new connective tissue in the root canal,and a large number of immature fibroblast-like cells and small vessels were seen to be gathered in the apical foramen,and there was no tendency for cells to enter the root canal. CXCR4+ cells were found in the root canals of N group and N+SDF-1 group,and a small amount of CXCR4+ cells were found in N+antiSDF-1 group. Conclusion: SDF-1/CXCR4 signal axis is involved in tissue regeneration and promotes angiogenesis by homing chemotactic cells into the root canal in dental pulp revascularization.
