非经典MHCⅠ类分子HLA-G基因多态性与广西肝癌家族聚集性的相关性研究

【摘要】 目的:探讨非经典组织相容性复合体(MHC)Ⅰ类分子人类白细胞抗原(HLA)-G基因多态性与广西肝癌家族聚集性的相关性。方法:采用病例对照研究设计,收集广西地区140例来自肝癌家族的家庭成员作为实验组,同期选择相同性别、年龄±5岁、相同民族、相同生活环境、相同生活习惯的140例无癌家族家庭成员作为对照组,通过聚合酶链反应(PCR)结合测序法检测HLA-G基因多态性。结果:实验组-14 bp/-14 bp基因型频率分布高于对照组,+14 bp/+14 bp、+14bp/-14 bp低于对照组(P<0. 05);实验组-14 bp等位基因分布高于对照组,+14 bp分布低于对照组。HLA-G+3142C/G位点、+3187A/G位点和HLA-G*0105N多态性分析显示,两组基因型和等位基因的分布比较,差异均无统计学意义(均P>0. 05)。结论:HLA-G基因14 bp插入/缺失多态性可能与肝癌家族聚集性有关。

【Abstract】 Objective: To investigate the correlation between the non-classical MHC class Ⅰ molecule HLA-G polymorphism and the familial aggregation of liver cancer in Guangxi. Methods: According to a casecontrol study,total of 140 patients of liver cancer families in Guangxi were selected as the experimental group,and 140 patients that without family history of liver cancer but of the same sex,age±5 years,same ethnicity,same living environment and same living habits were selected as a control group. HLA-G polymorphism was detected by polymerase chain reaction(PCR)and sequencing. Results: The 14bp insertion/deletion genotype analysis of the HLA-G gene found that the frequency distribution of the-14bp/-14bp genotype in the experimental group was significantly higher than that in the control group,while those of the +14bp/+14bp and +14bp/-14bp genotypes in the experimental group were significantly lower than the control group(all P<0. 05);the distribution of the-14bp allele in the experimental group was higher than that in the control group,and the distribution of + 14bp was lower than in the control group,and the differences were statistically significant(P<0. 05). Polymorphism analysis at HLA-G + 3142C/G locus,+ 3187A/G locus and HLA-G*0105N showed no significant difference in the distribution of genotypes and alleles between the two groups(all P>0. 05). Conclusion: The 14bp insertion/deletion polymorphism of HLA-G gene may be related to the familial aggregation of liver cancer.
