美洲大蠊提取物柔性脂质体凝胶的制备及其性能研究

【摘要】 为提高美洲大蠊提取物(PAE)的皮肤透过性及生物利用度,将其包封为柔性脂质体,并负载于凝胶基质上。利用乙醇注入法,将有机相与含有PAE的水相混合,制备了美洲大蠊提取物柔性脂质体(PAE-ULIP),对PAE-ULIP的形态、粒径、包封率等各项理化性质进行测定与表征,其中PAE-ULIP包封率相较于普通脂质体提升了4.6%。进一步以卡波姆-940为基质,丙二醇(1,2)为辅剂,制备了PAE-ULIP凝胶,对该凝胶皮肤透过性能进行测定,其24 h单位面积累积释放量为(690±38)μg×cm-2。结果表明,所制备的PAE-ULIP凝胶具有良好的皮肤透过性能、体外释药性能及抗炎活性。

【Abstract】 In order to improve the transdermal permeation and bioavailability of the periplaneta americana extract(PAE), ultradeformable liposome loaded on a gel matrix was applied for encapsulation. The organic phase was mixed with the aqueous phase containing PAE by ethanol injection to prepare the periplaneta americana extract ultradeformable liposome(PAE-ULIP). The physicochemical properties of PAE-ULIP, such as structure, size and encapsulation efficiency were measured. The encapsulation efficiency of PAE-ULIP was4.6% higher than that of normal liposomes. Furthermore, PAE-ULIP gel was prepared with carbomer-940 as matrix and 1,2-propanediol as adjuvant. The in vitro release characteristics of the gel were measured, and the cumulative release of 24 h was(690 ± 38) μg×cm-2. The results show that the prepared PAE-ULIP gel has good transdermal properties and in vitro release characteristics, and it also has certain anti-inflammatory activity.
