鸢尾生香菌的分离鉴定及鸢尾酮的生物合成

【摘要】 针对传统植物提取法生产鸢尾酮过程中存在的生产周期长、产物得率低、生产成本高等问题,提出以新鲜根粉为原料利用微生物发酵生产鸢尾酮的方法。从鸢尾根茎中筛选得到一株产香细菌,通过细菌16S rDNA基因序列分析确定为简单芽孢杆菌(Bacillus simplex strain)。使用甘油混合培养基,添加终浓度5 g×L-1的新鲜根粉为底物,在28℃,200 r×min-1条件下,发酵24 h顺式α-鸢尾酮摇瓶产量达到315.95 mg×kg-1。10 L发酵罐放大培养结果表明18 h后顺式α-鸢尾酮产量最高可达到1 037.65 mg×kg-1,是摇瓶发酵产量的3.28倍。利用微生物发酵生产鸢尾酮,大幅度提高了鸢尾酮的生产效率,对促进鸢尾酮生物合成的工业化应用具有重要意义。

【Abstract】 Traditional plant extraction for irone production has problems of long period, low yield and high cost. A method for irone production using fresh root powder as raw material by microbial fermentation was proposed. A bacterium producing aroma was screened from iris rhizome and identified as Bacillus simplex strain by 16 S rDNA sequence analysis. Using mixed media including glycerol and adding 5 g×L-1 fresh root powder as substrate, the fermentation production of cis-α-irone had a yield of 315.95 mg×kg-1 in shaking flask for 24 h at 28 ℃ and 200 r×min-1. The results in 10 L fermentor show that the maximum yield of cis-α-irone reaches 1 037.65 mg×kg-1 after 18 h, which is 3.28 times than that of shaking flask fermentation. The production of irone by microbial fermentation can greatly improve production efficiency, which is helpful for industrialization of irone biosynthesis.
