傅里叶红外光谱法在锁阳多糖提取工艺优化中的应用

【摘要】 优化提取工艺流程和参数通常是在单因素试验基础上,按照正交试验或响应面法设计实验方案,工作量大。将红外光谱技术应用到提取工艺单因素试验过程中,对锁阳多糖和锁阳药渣进行了研究,以期探讨一种快速辅助优化提取工艺的新方法。结果显示,锁阳多糖第1次提取液中含有油脂、蛋白等成分,不合并;提取3次后锁阳药渣中可提取多糖类成分已基本提取完全;酶用量1.5%时酶解作用充分,提取制备的锁阳多糖品质好。傅里叶红外光谱法可以快速辅助确定提取次数和酶用量的最佳水平,为下一步方案设计简化了影响因素,也为简化工艺流程提供了依据。

【Abstract】 It took heavy workload on optimization of extraction process, because of orthogonal test or RSM after one single factor experiment. In order to establish a new simple and rapid method for optimum extracting process, FTIR was used to detect CSP and CS residue in the single factor experiment. The results showed that the first CSP extract should not be merged due to its lipid and protein components. There was no need to extract more than three times, since the polysaccharides in CS residue had been completely extracted. When the enzyme concentration was 1.5%, the enzymatic hydrolysis was sufficient, and the CSP was of good quality. FTIR can provide quick determination for optimal extraction times and enzyme concentration, which simplified the factors for the orthogonal or RSM design, and also provide the basis for simplifying the extraction process.
