基于网络药理学的加减甘露消毒丹治疗RSV毛细支气管炎的分子作用机制研究

【摘要】 目的:探索加减甘露消毒丹治疗呼吸道合胞病毒(respiratory syncytial virus,RSV)毛细支气管炎的分子作用机制。方法:通过数据库联用与文献挖掘检索加减甘露消毒丹主要活性成分及其作用靶点、RSV毛细支气管炎相关的靶蛋白,使用Cytoscape 3.2.1软件作蛋白质相互作用网络,结合网络特征分析网络关键靶点,并将其投射到KEGG与GO数据库中进行富集与特征分析。结果:得到127个活性成分、158个成分作用靶点、38个疾病靶蛋白,经网络药理学分析得到推测其作用机制可能与B细胞与T细胞、TOLL样受体信号通路等有关。结论:甘露消毒丹中木樨草素等多种有效化合物可能通过NF-κB通路、B细胞与T细胞、TOLL样受体等途径发挥对RSV毛细支气管炎的治疗作用。

【Abstract】 Objective: To explore molecular mechanism of modified Ganlu Xiaodu pills in the treatment of RSV bronchiolitis. Methods: The combination of the databases and literature mining were used to search the main active ingredients and its targets of modified Ganlu Xiaodu pills, the target proteins related to RSV bronchiolitis,Cytoscape 3.2.1 software was used to construct protein-protein interaction network, network key targets were analyzed according to the characteristics of network, and projected to KEGG and GO database for enrichment and feature analysis. Results: All 127 active ingredients, 158 target compounds and 38 disease target protein were obtained, its mechanism might be related to B cells, T cells and Toll-like receptor signaling pathway and others through the analysis of network pharmacology. Conclusion: Luteolin and other effective compounds contained in Ganlu Xiaodu pills could play the therapeutic role in the treatment of RSV bronchiolitis, which might work through NF-κB pathway, B cells, T cells and Toll-like receptor signaling pathway.
