肤悦康洗剂对湿疹豚鼠的治疗作用及其免疫调节机制

【摘要】 目的:探讨肤悦康洗剂对湿疹豚鼠的治疗作用及其免疫调节机制。方法:将健康雌性豚鼠36只随机分为6组,每组6只。正常组于豚鼠脱毛处外涂100μL丙酮,其余各组在脱毛处外涂100μL 0.5%2,4-二硝基氯苯(dinitrochlorobenzene,DNCB),1周1次,共3次,诱发建立豚鼠湿疹模型。用浸泡在生理盐水中的纱布敷于模型组豚鼠脱毛处。阳性药物组外用哈西奈德溶液。肤悦康洗剂低、中、高剂量组分别用浸泡不同剂量肤悦康洗剂的纱布外敷于模型组豚鼠脱毛处。3周后观察各组豚鼠背部皮肤状态并评分;使用流式细胞仪检测各组豚鼠脾脏组织中细胞表面抗原CD4及细胞因子干扰素γ(interferon γ,IFN-γ)、白细胞介素4(interleukin-4,IL-4)、转化生长因子β(transforming growth factor-β,TGF-β)表达情况;使用酶联免疫吸附法(ELISA)检测各组豚鼠血清中细胞因子IL-2、IL-4、IFN-γ、IL-17、TGF-β表达。结果:与正常组相比,模型组豚鼠皮肤出现红斑、肿胀、糜烂、浸润等湿疹样皮损症状,皮损评分增高。阳性药物组及肤悦康洗剂各剂量组豚鼠皮损症状不同程度恢复。与模型组相比,阳性药物组及肤悦康洗剂各剂量组血清中IL-4、IL-17和TGF-β浓度降低,IL-2和IFN-γ浓度增加。阳性药物组及肤悦康洗剂各剂量组CD4~+细胞中IFN-γ、TGF-β表达增加,IL-4表达降低。肤悦康洗剂高剂量调节效果最明显。结论:肤悦康洗剂治疗豚鼠慢性湿疹疗效确切,其机制可能与其调节免疫功能作用有关。

【Abstract】 Objective: To explore the therapeutic effect of Fuyuekang lotion on eczema guinea pig model and its implicated mechanism on regulating immune function. Methods: All 36 healthy female guinea pigs were randomly divided into six groups, six each group. The other groups were prepared into eczema models by applying 100 μL 0.5% 2, 4-dinitrochlorobenzene(DNCB) to the depilation site, once a week, for three times, while the normal group were applied by 100 μL acetone. The model group accepted applying the gauze soaked in normal saline to the depilated area. Positive medicine group were smeared with Halcinonide solution. Low, moderate and high doses groups were applied with the gauze soaked in different doses of Fuyuekang lotion to the depilated area respectively.Back skin conditions of guinea pigs in different groups were observed and scored in three weeks; flow cytometry was used to detect the expressions of CD4, IFN-γ, IL-4 and TGF-β in spleen tissue; ELISA method was adopted to detect the expressions of IL-2, IL-4, IFN-γ, IL-17 and TGF-β in the serum of guinea pig. Results: Compared with the normal group, the skin of guinea pig in the model group manifested eczema-like lesions such as erythema swelling,erosion, infiltration and others, skin lesion score increased notably. Skin lesion symptoms of positive medicine group, different doses groups of Fuyuekang lotion recovered to different degrees. Compared with the model group,the concentrations of IL-4, IL-17 and TGF-β reduced in positive medicine group, different doses groups of Fuyuekang lotion, the concentrations of IL-2 and IFN-γ increased. The expressions of IFN-γ and TGF-β increased and the expressions of IL-4 lowered in CD4~+ cells in positive medicine group and different doses groups of Fuyuekang lotion. High dose of Fuyuekang lotion showed the most notable reguation effects. Conclusion: Fuyuekang lotion could obtain definite clinical effects in the treatment of guinea pig with chronic eczema, and its mechanism might be related to regulating immunity.
