针刺不同穴位对慢性炎症痛模型大鼠血清IL-4及IL-10的影响
Effects of Acupuncture at Different Acupoints on IL-4 and IL-10 in Rat Model with Chronic Inflammatory Pain
【摘要】 目的:观察针刺不同穴位对佐剂性关节炎模型大鼠血清中白细胞介素4(interleukin-4,IL-4)和白细胞介素10(interleukin-10,IL-10)水平的影响,并就其机制进行探讨。方法:将40只健康成年雄性Wistar大鼠随机分为正常对照组、佐剂模型组、佐剂关元组、佐剂针百会组、佐剂针足三里组,每组8只。除正常对照组外于每只大鼠右后足掌皮下注射佐剂0.1 m L造模,造模成功后佐剂针关元组、佐剂针百会组、佐剂针足三里组分别选取关元、百会、足三里穴进行针刺,进针后接韩式电针仪断续波,每次20 min,隔日治疗1次。观察大鼠体质量及一般状况,并于治疗后24、48、72 h分别观察局部足爪肿胀情况,同时于治疗72 h后测定血清IL-4、IL-10水平。结果:与正常对照组相比,佐剂模型组、佐剂针刺各组大鼠血清IL-4水平均呈升高趋势,其中以佐剂针百会组升高较为明显,差异有统计学意义(P<0.05);与佐剂模型组相比,佐剂针刺各组大鼠血清IL-4水平均呈升高趋势,其中佐剂针百会组显著升高,差异有统计学意义(P<0.05)。佐剂针刺各组中佐剂针关元组大鼠血清IL-4水平最低,佐剂针足三里组次之,而佐剂针百会组最高。与正常对照组相比,佐剂模型组和佐剂针关元组大鼠血清IL-10水平均降低,差异有统计学意义(P<0.01);佐剂针足三里和佐剂针百会组血清IL-10水平呈升高趋势,其中佐剂针足三里组与正常对照组相比,差异有统计学意义(P<0.05)。与佐剂模型组相比,佐剂针关元组大鼠血清IL-10水平降低,差异有统计学意义(P<0.05);佐剂针足三里和针百会组血清IL-10水平均明显升高,差异有统计学意义(P<0.01),其中佐剂针足三里组升高最为明显。佐剂各穴位组中佐剂关元组大鼠血清IL-10水平最低,佐剂百会组次之,而佐剂足三里组最高。结论:针灸佐剂性关节炎大鼠可以上调IL-4、IL-10水平,从而增强细胞因子的抗炎效应,但不同的穴位和方法作用于机体会产生不同的疗效。
【Abstract】 Objective: To observe the effects of acupuncture at different acupoints on the levels of IL-4 and IL-10 in the serum of rat model with adjuvant arthritis, and discuss its mechanism. Methods: All 40 healthy male Wistar rats were randomized into normal control group, adjuvant model group, the group of adjuvant and acupuncture at Guanyuan(CV4, RN4), the group of adjuvant and acupuncture at Baihui(DU20) and the group of adjuvant and acupuncture at Zusanli(ST36), eight rats each group. Except normal control group, the rat models were established by subcutaneous injection of adjuvant, 0.1 mL, into the palm of the right hindfoot, after establishing the models successfully, the group of adjuvant and acupuncture at Guanyuan(CV4, RN4), the group of adjuvant and acupuncture at Baihui(DU20) and the group of adjuvant and acupuncture at Zusanli(ST36) chose needling at Guanyuan, Baihui and Zusanli respectively, after needling insertion, they were connected with intermittent wave of Korean electroacupuncture instrument, 20 minutes each time, once every other day. To observe body mass and general situations of the rats, to observe local paw swelling after treating for 24, 48 and 72 hours respectively,simultaneously to detect the levels of IL-4 and IL-10 72 hours after treating. Results: Compared with normal control group, the levels of IL-4 in adjuvant model group and different groups of adjuvant needling were on the rise, among them, the increase of the group of adjuvant and acupuncture at Baihui was more notable, and the difference had statistical meaning(P <0.05); compared with adjuvant model group, the levels of IL-4 in different groups of adjuvant needling were on the rise, among them, the group of adjuvant and acupuncture at Baihui increased notably,and the difference had statistical meaning(P<0.05). Among these groups, the levels of IL-4 of the group of adjuvant and acupuncture at Guanyuan were the lowest, the group of adjuvant and acupuncture at Zusanli the next, while the group of adjuvant and acupuncture at Baihui the highest level. Compared with normal control group, the levels of IL-10 in the rats of adjuvant model group and the group of adjuvant and acupuncture at Guanyuan lowered, and the difference showed statistical meaning(P <0.01); the levels of IL-10 in the group of adjuvant and acupuncture at Zusanli and the group of adjuvant and acupuncture at Baihui were increasing, the difference was statistically significant when the group of adjuvant and acupuncture at Zusanli was compared with normal control group(P<0.05).Compared with adjuvant model group, the levels of IL-10 reduced in the group of adjuvant and acupuncture at Guanyuan, and the difference had statistical meaning(P<0.05); the levels of IL-10 rose in the group of adjuvant and acupuncture at Zusanli and the group of adjuvant and acupuncture at Baihui rose notably, and the difference had statistical meaning(P<0.01), among them, the increase of the group of adjuvant and acupuncture at Zusanli was the most notable. The levels of IL-10 were the lowest in the group of adjuvant and acupuncture at Guanyuan, the group of adjuvant and acupuncture at Baihui the next, while the levels of the group of adjuvant and acupuncture at Zusanli the highest. Conclusion: Acupuncture could up regulate the levels of IL-4 and IL-10, therefore to enhance anti-inflammatory effects of cytokines, but different acupoints and method acting on the body could produce different clinical effects.
