油茶肉质果多糖诱导糖尿病小鼠肝细胞生产胰岛素的实验研究

【摘要】 目的:初步探讨油茶肉质果多糖诱导糖尿病小鼠肝细胞生产胰岛素及其可能的机制。方法:建立糖尿病小鼠模型,采用250 mg·kg-1油茶肉质果多糖灌胃糖尿病小鼠3个月后,取其肝脏,然后通过免疫荧光、免疫组化及Western blot分析油茶肉质果多糖是否能够诱导糖尿病小鼠肝细胞生产胰岛素。结果:油茶肉质果多糖能够诱导糖尿病小鼠肝细胞生产胰岛素,且能使糖尿病小鼠肝脏中转录因子胰十二指肠同源盒基因(PDX-1)的蛋白表达上调。结论:油茶肉质果多糖可以诱导糖尿病小鼠肝细胞生产胰岛素。

【Abstract】 Objective :To investigate whether polysaccharides from the fleshy fruit of Camellia oleifera Abel can induce the differentiation of mature hepatocytes into insulin secreting cells and the potential mechanism.Methods :Diabetic mouse model was established,and the diabetic mice were administered orally with 250 mg·kg-1 polysaccharides from the fleshy fruit of Camellia oleifera Abel for 3 months.After that,the animals were sacrificed and their livers were immediately removed.And the livers were analyzed by immunofluorescence,immunohistochemistry,and Western blot to determine whether polysaccharides from the fleshy fruit of Camellia oleifera Abel could induce the differentiation of hepatocytes of diabetic mice into insulin-secreting cells.Results:The results showed that polysaccharides from the fleshy fruit of Camellia oleifera Abel could induce hepatocytes of diabetic mice to differentiate into insulin-secreting cells,and could upregulate the protein expression of PDX-1 in the liver of diabetic mice.Conclusion :Polysaccharides from the fleshy fruit of Camellia oleifera Abel can induce the insulin production of hepatocytes
