龙须藤总黄酮对糖尿病大鼠肾脏中NF-κB信号通路的影响
Bauhinia championii(Benth.) Benth flavones alleviates streptozotocin-induced diabetic kidney disease of diabetic rats via inhibiting NF-κB signal pathway
【摘要】 目的:探讨龙须藤总黄酮对链脲佐菌素诱导的糖尿病大鼠肾脏中NF-κB信号通路的影响。方法:通过免疫组化、免疫荧光和Western Blot的方法研究龙须藤总黄酮对糖尿病大鼠肾脏中NF-κB信号通路是否有抑制作用。结果:免疫组化和免疫荧光结果均发现糖尿病大鼠肾皮质中P65在细胞核中的分布较多,而给予龙须藤总黄酮治疗后P65在细胞核中的分布与糖尿病组比较明显减少;分离细胞浆蛋白和细胞核蛋白,然后采用Western Blot检测P65在细胞浆和细胞核中的分布,结果发现,糖尿病组大鼠肾皮质中P65在细胞核中的分布比在细胞浆中的分布多,而采用龙须藤总黄酮治疗后P65在细胞核中的分布明显减少,抑制了P65的核转位。结论:龙须藤总黄酮能够抑制糖尿病大鼠肾脏中NF-κB信号通路的激活。
【Abstract】 Objective :To investigate the effect of Bauhinia championii(Benth.)Benth flavones on NF-κB signal pathway in the kidney of diabetic rats.Methods:It was investigated that whether the total flavonoids of Bauhinia championii(Benth.)Benth could inhibit the NF-κB signaling pathway in the kidneys of diabetic rats by immunohistochemistry,immunofluorescence and Western Blot.Results :Immunohistochemistry and immunofluorescence results showed that the distribution of P65 in the nucleus of the kidney cortex of the flavonoids treated diabetic rats was significantly reduced compared with that of the diabetic rats;The cytoplasmic protein and nuclear protein were separated,and then the distribution of P65 in the cytoplasm and nucleus was detected by Western Blot. It was found that the distribution of P65 in the nucleus of the kidney cortex of the diabetic group was more than that in the cytoplasm. The distribution of P65 in the cell nucleus was significantly reduced after the treatment with the total flavonoids from Bauhinia championii(Benth.)Benth,these results indicated that flavonoids from Bauhinia championii(Benth.)Benth,could inhibit the nuclear translocation of P65.Conclusion : Bauhinia championii(Benth.)Benth flavones could alleviate streptozotocin-induced diabetic kidney disease of diabetic rats via inhibiting NF-κB signal pathway.
