大鼠结肠癌模型H-ras和KISS-1表达与其病理分期及复发转移的关系
Relationship between expression of H-ras and KISS-1 and pathological stage and recurrence and metastasis of colon cancer in rats
【摘要】 目的探讨H-ras、KISS-1表达与大鼠结肠癌病理分期及复发转移的关系。方法选取60只SD大鼠均颈背部皮下注射二甲肼,每周注射1次,连续注射5周,构建结肠癌大鼠模型,并给予奥沙利铂15 mg/kg,腹腔注射,每周2次,连续给药8周。免疫组织化学法检测大鼠结肠癌组织H-ras、KISS-1蛋白表达;Western blot法和RealTime PCR检测结肠癌组织H-ras、KISS-1蛋白含量和mRNA含量的表达;并分析H-ras、KISS-1表达与大鼠结肠癌病理分期及复发转移的相关性。结果与治疗前相比,大鼠结肠癌组织内H-ras平均光密度、H-ras蛋白表达及H-ras mRNA含量显著降低;KISS-1平均光密度、KISS-1蛋白表达及KISS-1 mRNA含量显著升高(P<0.05)。H-ras和KISS-1阳性表达和结肠癌的淋巴结转移、浸润深度、分化程度、有无远处转移、Dukes分期和TNM分期具有明显的相关性(P<0.05)。结论大鼠结肠癌组织中H-ras和KISS-1异常表达与肿瘤转移转移密切相关,可作为预测结肠癌分期和淋巴结转移的分子生物学靶点。
【Abstract】 Objective To investigate the relationship between the expression of H-ras and KISS-1 and the pathological stage and recurrence and metastasis of colon cancer in rats.Methods Forty SD rats were injected subcutaneously with dimethylhydrazine,once a week for 5 weeks,and a colon cancer rat model was established.Oxaliplatin 15 mg/kg was given intraperitoneally,weekly.2 times,continuous administration for 8 weeks.The expression of H-ras and KISS-1 protein in rat colon cancer tissues was detected by immunohistochemistry.The expression of H-ras and KISS-1 protein and mRNA in colon cancer tissues were detected by Western blot and Real-Time PCR.Correlation between the expression of-ras and KISS-1 and the pathological stage and recurrence and metastasis of colon cancer in rats.Results Compared with pre-treatment,H-ras mean optical density,H-ras protein expression and H-ras mRNA content in rat colon cancer tissues were significantly decreased;KISS-1 average optical density,KISS-1 protein expression and KISS-1 mRNA content was significantly increased(P<0.05).The positive expression of H-ras and KISS-1 was significantly correlated with lymph node metastasis,depth of invasion,degree of differentiation,presence or absence of distant metastasis,Dukes stage and TNM stage(P<0.05).Conclusion The abnormal expression of H-ras and KISS-1 in rat colon cancer tissues is closely related to tumor metastasis and metastasis,and can be used as a molecular biological target for predicting colon cancer stage and lymph node metastasis.
