乐伐替尼抑制VEGF/MAPK/NF-κB治疗结肠癌及对患者T淋巴细胞亚群的影响
Effect of levantinib on VEGF/MAPK/NF-κB in the treatment of colon cancer and its effect on t lymphocyte subsets
【摘要】 目的探讨乐伐替尼抑制VEGF/MAPK/NF-κB治疗结肠癌及对患者T淋巴细胞亚群的影响。方法选取2016年1月-2018年10月本院收治的100例结肠癌患者,患者接受口服乐伐替尼24mg/d直至疾病恶化和出现无法耐受的不良反应。评价药物使用疗效;观察患者T淋巴细胞亚群变化;采用免疫组织化学法检测患者癌组织中VEGF蛋白的表达;采用Western blot法检测甲状腺癌癌组织中VEGF、p-p38、p-JNK、p-ERK、NF-κB和Caspase3蛋白的表达。结果乐伐替尼治疗结肠癌总有效率为60.00%。结肠癌患者应用乐伐替尼治疗后,患者CD3~+、CD4~+、CD4~+/CD8~+表达显著上升,而CD8~+表达水平则显著下降(P<0.05);癌组织内VEGF蛋白平均光密度含量明显下降(P<0.05);VEGF、p-ERK蛋白含量显著降低,p-p38、p-JNK、NF-κB和Caspase 3蛋白含量显著升高(P<0.05)。结论乐伐替尼抑制VEGF/MAPK/NF-κB信号通路治疗结肠癌,并提高患者T淋巴细胞数量,改善患者免疫功能。
【Abstract】 Objective To investigate the effect of levabinib on the inhibition of VEGF/MAPK/NF-κB in colon cancer and T lymphocyte subsets in patients.Methods One hundred colon cancer patients admitted to our hospital from January 2016 to October 2018 were enrolled.The patients received oral levolutini 24 mg/d until the disease worsened and the intolerable side effects were observed.Evaluate the efficacy of drug use;observe the changes of T lymphocyte subsets in patients;detect the expression of VEGF protein in cancer tissues by immunohistochemistry;detect VEGF,p-p38,p-JNK,p in thyroid carcinoma by Western blot-expression of ERK,NF-κB and Caspase 3 proteins.Results The total effective rate of levabinib in the treatment of colon cancer was 60.00%.After treatment with lovatinib in colon cancer patients,CD3~+,CD4~+,CD4~+/CD8~+ expression increased significantly,while CD8~+ expression decreased significantly(P<0.05).The average optical density of VEGF protein in cancer tissues decreased significantly(P<0.05).The contents of VEGF and p-ERK protein were significantly decreased,and the contents of p-p38,p-JNK,NF-κB and Caspase 3 were significantly increased(P<0.05).Conclusion Levantinib inhibits VEGF/MAPK/NF-κB signaling pathway in the treatment of colon cancer,and increases the number of T lymphocytes in patients,improving the immune function of patients.
