miR-497对非小细胞肺癌患者的诊断价值

【摘要】 微小RNA(miRNA)是一类非编码小分子RNA,长度为18~25个核苷酸,通过互补的碱基与特定m RNA的3’-非翻译区(3’-UTR)结合,进而使靶m RNA发生降解或影响其翻译,用于转录后水平调控基因的表达。不同或者相同的miRNA在不同肿瘤中表现出作用不一样,且涉及肿瘤的每个发展阶段。本研究旨在探讨miR-497在非小细胞肺癌患者组织中的表达情况及临床应用价值。方法:采用Real-time PCR法检测10例非小细胞肺癌患者组织及癌旁,比对其表达差异,同时绘制ROC曲线判断其诊断价值。结果:非小细胞肺癌患者中miR-497在肺癌组织中表达下调,可能与非小细胞肺癌的发生密切相关,进一步分析得出,miR-497可能会成为诊断非小细胞肺癌的早期诊断指标。

【Abstract】 Micrornas(mirnas) are a class of non-coding small RNA with a length of 18 to 25 nucleotides that bind to the 3 ’-untranslated region(3’-utr) of specific m RNA through complementary bases, thereby degrading the target mRNA or affecting its translation, and are used to regulate gene expression at the post-transcriptional level. Different or identical mirnas have different roles in different tumors and are involved in each stage of tumor development. The purpose of this study was to investigate the expression and clinical value of mir-497 in tissues of patients with non-small cell lung cancer. Methods: real-time PCR was used to detect the tissues and periamcinoma of 10 patients with non-small cell lung cancer(NSCLC). Results: mir-497 expression was down-regulated in lung cancer tissues in patients with non-small cell lung cancer, which may be closely related to the occurrence of non-small cell lung cancer. Further analysis showed that mir-497 might be an early diagnostic indicator for non-small cell lung cancer.
