‘东红’猕猴桃组织培养体系的建立

【摘要】 ‘东红’猕猴桃是从‘红阳’猕猴桃实生苗中选育而来,具有口感细腻、风味浓郁、早熟、耐贮等特点,具有较高的生产栽培价值。以1年生茎段为外植体,建立‘东红’猕猴桃不定芽再生体系。结果表明,茎段可以在1/2 MS+2.0 mg·L-16-BA+0.5 mg·L-1NAA+20 g·L-1蔗糖的培养基上萌发出不定芽;最佳增殖培养基为MS+1.5 mg·L-16-BA+0.1 mg·L-1NAA,增殖率为6.78;最适不定芽生根培养基为1/2 MS+1.0~1.5 mg·L-1IBA,生根率均高于90%;在泥炭土∶田园土=2∶1组成的混合基质中,再生苗移栽成活率为93%。为猕猴桃优良品种的保存和推广奠定基础。

【Abstract】 ‘Donghong’ kiwifruit cultivar was selected from seedlings of Actinidia chinensis ‘Hongyang’. It showed delicate taste,rich flavor, early maturing, long storage, and etc., having good value in production and cultivation. The adventitious bud regeneration system of ‘Donghong’ Kiwifruit was established by using annual stem segment as explants. The result showed that adventitious buds were germinated on 1/2 MS+2.0 mg·L-16-BA+0.5 mg·L-1 NAA+20 g·L-1. The most suitable medium for shoot multiplication was MS+1.5 mg·L-16-BA+0.1 mg·L-1 NAA and multiplication rate was up to 6.78. The optimum medium for adventitious bud rooting was1/2 MS+1.0~1.5 mg·L-1 IBA, and rooting rate was up to 90%. The survival rate was as high as 93% in the mixture substrate of peat soil and pastoral soil(2:1). The results laid a sound foundation for preservation and promotion of fine varieties.
