猪流行性腹泻病毒陕西地方株的分离鉴定及序列分析
Isolation,Identification and Sequence Analysis of Local PEDV Strains in Shaanxi
【摘要】 为了解陕西地区猪流行性腹泻病毒(PEDV)的遗传进化方向及为疫苗的研发奠定基础,采集陕西省部分地区腹泻仔猪的肠道及内容物,利用Vero细胞进行病原分离,扩增M基因和N基因序列并测序。结果显示病毒盲传5代,细胞出现收缩变圆直至脱落的细胞病变;序列分析结果表明,分离出的PEDV陕西地方株与其他参考毒株M基因核苷酸序列相似性为97.8%~99.7%,氨基酸序列相似性为97.3%~100%;N基因核苷酸序列相似性为95.7%~99.8%,氨基酸序列相似性为99.3%~100%。PEDV陕西分离株与CV777等经典株的亲缘关系较远,与变异株的亲缘关系较近。表明成功分离出PEDV陕西地方株,且均为变异株。
【Abstract】 To understand the genetic evolution direction of porcine epidemic diarrhea virus(PEDV)in Shaanxi and lay the foundation for vaccine research,the intestinal tract and contents of diarrhea piglets in some areas of Shaanxi province were collected,and Vero cells were used for pathogen isolation.The M gene and N gene were amplified and sequenced.The results showed that the virus was passaged blindly for five generations,the cells showed shrinkage and rounding until the detached cytopathic lesions.The sequence results showed that the nucleotide similarity in the M gene between the isolated PEDV Shaanxi local strains and other reference strains was 97.8%-100%,the amino acid similarity was 97.3%-100%;the nucleotide similarity in the N gene was 95.7%-99.8%,and the amino acid similarity was 99.3%-100%.The PEDV Shaanxi isolates are far from the classical strains such as CV777,and are closely related to the mutant strainsI.tindicatedthatthePEDVShaanxilocal strains were successfully isolated and all were mutants.
