没食子酸对南方鲇红细胞抗氧化酶和表面形貌的影响

【摘要】 为了解不同浓度没食子酸(GA)对南方鲇(Silurus meridionalis)红细胞抗氧化酶和表面形貌的影响,在(27±0.5)℃时用南方鲇亚成体血液制备1%红细胞悬液,添加没食子酸,检测超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)的活性和丙二醛(MDA)含量,并且观察红细胞相应的溶血和表面形貌的变化。结果显示:溶血OD值与时间成正比关系,4 h溶血OD没有差异,24 h溶血OD先降后升;无论4 h还是24 h三种抗氧化酶均为先升高再降低的趋势,MDA先下降再升高的趋势,且24 h的变化趋势比4 h更加明显;红细胞表面形貌发现4 h时,除80 mg/L浓度组可以看到少数红细胞表面略有皱缩变形,其它组与对照组无差异,24 h时5、10、20 mg/L浓度组与对照组相比红细胞均无变化,40和80 mg/L浓度组红细胞表面损伤明显。结果表明:GA对健康南方鲇红细胞抗氧化酶的活性和表面形貌的影响是双向的,20 mg/L以下有抗氧化作用,超过20 mg/L促氧化作用。

【Abstract】 The oxidation and morphology of red blood cells(RBCs)from Silurus meridionalis in vitro exposure to gallic acid(GA)were reported.1% RBCs were incubated with 0 to 80 mg/L GA for 4 h and 24 h at(27±0.5)℃.The results showed that the hemolytic OD were direct proportion to time,no difference in 4 h and increased first and decreased later in 24 h.The superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GPx)decreased and malondialdehyde(MDA)increased slightly.Three enzymatic antioxidants activities increased initially and then decreased with concentrations.However,MDA decreased initially and then increased,which was more obvious in 24 h rather than in 4 h.The morphology had crimples and deformation at the 80 mg/L and no significance at 5 to 40 mg/L compared to the controls in 4 h.No significance was found at 5 to 20 mg/L and visible morphologiccal damage at 40 and 80 mg/L in 24 h.It showed that the biphasic concentration-dependent curves with differing degrees on the enzymatic antioxidants activities and morphology with GA of RBCs from S.Meridionalis and showed antioxidation below 20 mg/L and promoting oxidation above 20 mg/L.
