缺氧诱导因子2α在大鼠早期妊娠中的表达与调节

【摘要】 利用原位杂交、免疫组织化学、Western blot以及荧光定量PCR等试验技术,研究HIF-2α基因在大鼠早期妊娠1~9 d、激素处理、人工诱导蜕膜化及体外诱导人子宫基质细胞蜕膜化模型中表达情况。妊娠1~5 d子宫基质细胞中低表达;妊娠第6天在围绕着床位点子宫基质细胞中HIF-2α表达升高;妊娠第7~9天HIF-2α蜕膜中表达逐渐增强;24 h雌激素和孕酮处理HIF-2α在子宫中表达无变化。HIF-2α在人工诱导蜕膜化子宫中高表达。在体外诱导人子宫内膜基质细胞蜕膜化模型中,HIF-2α随诱导天数增加表达升高。体内体外试验结果表明,HIF-2α可能在早期妊娠蜕膜化过程中发挥作用。

【Abstract】 In situ hybridization, immunohistochemistry, Western blot, and Real-time PCR methods were used to detect the expression of HIF-2α gene in early pregnancy, hormone regulation, artificially induced decidualization in rats and in human decidual cells induced in vitro. Low expression of HIF-2αwas detected in uterine stromal cells from the 1 th to the 5 th day of pregnancy; expression of HIF-2αincreased in uterine stromal cells surrounding the implantation site on the 6 th day of pregnancy; on the 7 th to 9 th day of pregnancy, the expression of HIF-2α gradually increased in the decidual area. There was no change in HIF-2α expression in the uterus after 24 h of estrogen and progesterone treatment. HIF-2α was highly expressed in the uterus with artificially induced decidualization. In the human uterus of steroid hormone treatment model in vitro, the expression of HIF-2α increased with the increase of induction days.These in vitro and in vivo results suggested that HIF-2α might play a role in the decidualization of early pregnancy.
