鸡内源性白血病毒ev21与其侧翼区SNP位点的连锁关系
The linkage between chicken endogenous leukemia virus ev21 and the SNP locus in the cell flanking region
【摘要】 鸡内源逆转录病毒ev21的转录表达影响宿主机体健康。本试验通过PCR检测海兰褐、太行鸡、海兰灰和SPF鸡292份DNA样品的ev21阴阳性、羽型和性别,测序筛选ev21插入位点OS和US区的SNP位点,MboII酶切检测C>T突变的群体多样性,分析C>T突变与ev21整合的连锁关系;最后利用TRANSFAC?Professional预测C>T突变引起的转录因子变化。测序峰图发现,ev21插入位点上游278 bp处的C>T突变。OS和US共有610 bp酶切结果发现,快羽ev21阴性鸡为CC基因型,快羽ev21阳性鸡为CT基因型;慢羽除海兰灰和海兰褐祖代ev21阳性鸡为TT基因型外,其他均为CT基因型。OS特异1 036 bp酶切显示ev21整合的OS区全部为T单倍型,C>T突变造成转录因子结合变化。研究表明ev21整合的OS区和未整合的US区的C>T突变有5种单倍型,ev21整合与T等位基因连锁。
【Abstract】 The endogenous retrovirus ev21 in chickens has bad effects on the host health. A total of 292 blood DNA samples from four different breeds, including Hailan Brown progenitors and commercial stocks, Taihang chickens, Hailan Gray and SPF chickens were collected. The feathering type, gender and ev21 insertion situation was detected by PCR amplification. SNP was screened by sequencing the OS and US region of ev21 integration. Mbo II digestion was used to detect the population diversity of C>T mutation and the linkage between the mutation and ev21 integration was analyzed. The changes of transcription factors caused by the SNP were predicted by TRANSFAC?Professional. The linkage between ev21 integration and the C>T mutation at 278 bp upstream of the integration site was found from the peak figure of sequencing. From the digestion results of 610 bp for the OS and US common region, it was found that the genotype of the SNP in ev21-negative and fast-feathering chickens was CC type, and CT genotype in ev21-positive and fast-feathering chickens. The late-feathering chickens were CT genotype except for TT genotype in ev21-positive Hailan Brown progenitors and Hailan Gray chickens. The digestion experiment of 1 036 bp special in OS region showed that the OS region for ev21 integration was all T haplotype. The C>T mutation caused the combination changes of transcription factors. The experiments showed that the C>T mutation in OS region for ev21 integration and US region without ev21 had five haplotypes, and ev21 integration was linked with T allele.
