鲁氏不动杆菌降解高效氯氰菊酯条件的响应面优化

【摘要】 目的探讨鲁氏不动杆菌J-2降解氯氰菊酯的最优条件。方法以鲁氏不动杆菌J-2为对象,采用单因素法考察环境因素对鲁氏不动杆菌降解高效氯氰菊酯效果的研究,再采用Box-Behnken实验设计和响应曲面法(RSM)优化降解条件。结果单因素培养温度、底物浓度和装载量为影响菌株J-2降解高效氯氰菊酯的主要因素,响应面优化的最优条件下鲁氏不动杆菌J-2在72h对高效氯氰菊降解率达到81.5%,降解量可达9.78mg。结论响应面法优化菌株J-2降解高效氯氰菊酯的最优条件为:温度25℃、底物浓度300mg/L、初始pH 6.0,接种量9%,装载量40mL,本研究为菌株应用于去除或消减拟除虫菊酯农药污染的环境提供理论基础。

【Abstract】 Objective To explore the optimal conditions of degrading cypermethrin by Acinetobacter rouxii J-2.Methods Acinetobacter lwoffii J-2 was taken as the research object and the influence of enviromental factors on its effect of degrading beta-cypermethrin was studied by the single factor method.The degradation conditions were optimized by the Box-Behnken experimental design and response surface methodology(RSM).Results The main factors affecting the degradation of beta-cypermethrin were the three single factors including the culture temperature,substrate concentration and loading capacity.Under the optimal condition of the response surface,the degradation rate of beta-cypermethrin by Acinetobacter lwoffii J-2 was 81.5%and the degradation amount was 9.78 mg at 72 hours.Conclusion The optimal conditions of degrading beta-cypermethrin by Acinetobacter lwoffii J-2 are as follows:the temperature is 25℃,the substrate concentration is 300 mg/L,the initial pH value is 6.0,the inoculation amount is 9% and the loading capacity is 40 mL.The findings provide a theoretical basis for the application of Acinetobacter lwoffii J-2 in removing or reducing the pesticide pollution of pyrethroid.
