基于网络药理学的黄芪-丹参药对治疗胃癌前病变作用机制

【摘要】 目的:探讨黄芪-丹参药对治疗胃癌前病变(GPL)的有效活性成分及作用机制。方法:本研究通过TCMSP数据库选取黄芪-丹参药对中含有的289个化合物,筛选其活性成分及潜在靶点;同时,通过Genecards数据库挖掘GPL疾病靶点;结合化合物及疾病靶点,构建化合物-靶点网络、进行GO富集分析及KEGG通路富集分析。结果:黄芪-丹参药对中含有76个活性成分,对应GPL 93个靶点;获得17条GO生物过程相关条目与6条信号通路,涉及肿瘤信号通路、PI3k/Akt信号通路、Ras/MAPK信号通路等。结论:黄芪-丹参药对主要通过调控细胞代谢来治疗GPL,为阐明黄芪-丹参药对治疗GPL的作用机制、开展实验验证提供依据。

【Abstract】 Objective:To investigate the active ingredients and mechanisms of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines in treating gastric precancerous lesions (GPL).Methods:A network pharmacology approach was used to detect the 289 ingredients in Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines.By searching the TCMSP and Genecards database,the targets which combined the decoction and disease were obtained.Then,gene ontology (GO) enrichment and Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analyses were constructed to explore the therapeutic mechanism of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines.Results:Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines have 76 active compounds corresponding to 93 targets of GPL.In GO enrichment analysis,there were 17 GO terms related to biological process.In KEGG pathway enrichment analysis,there were 6 KEGG pathways,including tumor signal pathways,PI3k/AKT signal pathway,Ras/MAPK signal pathway and so on.Conclusion:The present work indicates that the Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines mainly focus on treating GPL by regulating metabolism,which provides a basis for clarifying the mechanism of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae couplet medicines in the treatment of gastric precancerous lesions and carrying out experimental verification.
