c-Met通过E-cadherin介导的EMT与宫颈癌进展的相关性研究
Correlation between the regulation of c-met in E-cadherin-induced EMT and cervical cancer progression
【摘要】 目的:分析c-met、E-cadherin在宫颈癌组织中表达水平的变化,与癌组织病理特征之间的相关性,探究c-met通过E-cadherin介导的上皮-间质转化(Epithelial mesenchymal transition,EMT)参与宫颈癌发生、发展的可能作用机制。方法:收集内蒙古自治区人民医院和包头医学院第一附属医院2015年3月至2017年3月收治的50例宫颈癌患者的肿瘤组织及癌旁组织,采用免疫组织化学法检测癌组织及癌旁组织中c-met和E-cadherin表达;采用配对t检验对比分析癌组织与癌旁组织中c-met、E-cadherin表达;应用Spearson线性相关检验分析c-met、E-cadherin表达水平与宫颈癌患者临床病理特征间的相关性;应用Kaplan–Meier和Log-rank检验统计分析c-met、E-cadherin表达对宫颈癌患者生存情况的影响。结果:宫颈癌组织中c-Met表达高于癌旁组织,E-cadherin表达低于癌旁组织;24个月随访结果显示,c-met表达越低,E-cadherin表达越高,宫颈癌患者生存率越高。结论:c-met在宫颈癌组织中表达上调,从而抑制E-cadherin表达,触发癌细胞的增殖、迁移与浸润,降低癌症患者生存率。
【Abstract】 Objective:To analyze the expression changes of c-met and E-cadherin in cervical cancer tissues and their correlation with the pathological features of cancer tissues and to explore the potential mechanism of c-met in cervical cancer genesis and progression via regulating E-cadherin induced EMT. Methods: Tumor tissues and the adjacent tissues were collected from 50 cases of cervical cancer patients admitted to People’s Hospital of Inner Mongolia Autonomous Region and The First Affiliated Hospital of Baotou Medical College from March 2015 to March 2017 and their c-met and E-cadherin expressions were examined by using immunohistochemical method; the C-Met and E-cadherin expressions of tumor tissues and the adjacent tissues were compared and analyzed by using the paired t-test; the correlation between c-met and E-cadherin expressions and cervical cancer patients’ clinical pathological features was analyzed by using Spearson linear correlation test; the impact of c-met and E-cadherin expressions on cervical cancer patients’ survival was analyzed by using the Kaplan-Meier and Log-rank tests. Results: The C-Met expression is higher in cervical cancer tissues than in the adjacent tissues, while the E-cadherin expression is lower than in the adjacent tissues. The 24-month follow-up showed that the lower the c-met expression was, the higher the E-cadherin expression was, and so was the cervical cancer patients’ survival rate. Conclusion: The upregulation of c-met expression can inhibit E-cadherin expression, trigger the proliferation, migration and infiltration of cancer cells, and thus reduce the patients’ survival rate.
