黑龙江省黄瓜棒孢叶斑病分布及病菌生物学特性

【摘要】 对黑龙江省保护地发生的黄瓜棒孢叶斑病的发生情况进行调查,对疑似该病的致病菌进行形态学及分子生物学鉴定,同时以鉴定出的黄瓜棒孢叶斑病菌菌株JMS-1fh为试验材料,采用菌落生长法和玻片法对病原菌的生物学特性进行测定。结果表明:黄瓜棒孢叶斑病菌的病原菌为多主棒孢霉(Corynespora cassiicola(Berk&Curt)Wei.),该病在黑龙江省哈尔滨、齐齐哈尔、牡丹江等多地均有发生。病原菌菌丝生长的最适温度为25~28℃,最适碳源为甘露醇,最适氮源为甘氨酸,最适pH为6~8,光照条件有利于菌丝的生长,菌丝致死温度为55℃、10 min,分生孢子致死温度为55℃、5 min,分生孢子在25℃时萌发率最高。

【Abstract】 The occurrence of cucumber target leaf spot in Heilongjiang Province was investigated,the pathogenic fungus suspected of the disease were identified by morphology and molecular biology,and the identified strains of Corydalis leaf spot were used as experimental materials.The biological characteristics of pathogenic fungus were determined by colony growth method and glass slide method.The occurrence of cucumber leaf spot disease was investigated in the protected area of Heilongjiang Province,and the hazard symptoms in detail was described.The pathogenic fungus of the disease were identified by morphology and molecular biology.At the same time,the biological characteristics of the pathogen were determined by colony growth method and slide method.The results showed that the pathogen of cucumber rod leaf spot was Corynespora cassiicola(Berk & Curt) Wei.. The disease occured in Harbin,Qiqihar,Mudanjiang and other places in Heilongjiang Province.The optimum temperature for mycelium growth of pathogenic fungus was 25-28 ℃.The pathogen could make use of various carbon and nitrogen sources,mannitol was the best carbon source and glycine was the best nitrogen source.The optimum pH of 6-8 and the light were favorable for the growth of mycelia.The lethal temperature of hyphal was 55 ℃,with treating 10 minutes,the lethal temperature of conidia was 55 ℃,with treating 5 minutes,and the highest germination rate of conidia was at 25 ℃.
