宫颈癌患者外周血小核仁RNA宿主基因1表达及临床意义
Expression of small nucleolar RNA host gene 1 in peripheral blood cells and its clinical significance in patients with cervical cancer
【摘要】 目的研究宫颈癌患者外周血细胞中小核仁RNA宿主基因1(SNHG1)表达情况,并分析SNHG1异常表达在宫颈癌中的临床意义。方法定量逆转录聚合酶链反应(RT-PCR)检测56例宫颈癌患者和30例健康受试者外周血,以及宫颈癌患者宫颈癌组织及癌旁组织中SNHG1 mRNA的相对表达量,并分析外周血中SNHG1的表达与宫颈癌组织中SNHG1的相关性,探讨SNHG1的表达与宫颈癌患者临床特征的关系,及其对患者预后的影响。以受试者工作特征(ROC)曲线和曲线下面积(AUC)评估SNHG1的表达对宫颈癌的诊断价值。结果宫颈癌患者外周血SNHG1 m RNA相对表达量明显高于对照组,宫颈癌组织中SNHG1 mRNA相对表达量明显高于癌旁组织,差异均有统计学意义(P<0.01)。ROC曲线结果显示,SNHG1的表达诊断宫颈癌的AUC为0.836(95%CI:0.748~0.924),差异有统计学意义(P<0.01)。以外周血SNHG1 mRNA相对表达量0.551为界,SNHG1高表达宫颈癌患者30例,SNHG1低表达宫颈癌患者26例,临床分期为Ⅰ~Ⅱ期、无淋巴结转移和术后无复发的宫颈癌患者外周血中SNHG1呈低表达,差异均有统计学意义(P<0.05)。Spearman相关分析结果显示,宫颈癌患者外周血SNHG1的表达与宫颈癌组织中SNHG1的表达呈正相关(r=0.785,P<0.01)。SNHG1高表达宫颈癌患者的总生存时间明显短于SNHG1低表达患者,差异有统计学意义(P<0.01)。结论宫颈癌患者外周血SNHG1的表达上调,是宫颈癌辅助诊断及预后评估的潜在分子标志物。
【Abstract】 Objective To investigate the expression level of small nucleolar RNA host gene 1(SNHG1) in peripheral blood cells of patients with cervical cancer,and analyze the clinical significance of its abnormal expression level in cervical cancer. Method Quantitative reverse transcription polymerase chain reaction(RT-PCR) was used to detect the mRNA expression in peripheral blood cells from 56 patients with cervical cancer and 30 healthy subjects,as well as the relative expression of SNHG1 mRNA in either cervical cancer tissues or adjacent tissues of cervical cancer patients.The correlation between the expression of SNHG1 in peripheral blood and cervical cancer tissue,and its expression with the clinical features,as well as the impact on the prognosis of cervical cancer patient,were also analyzed.The receiver operating characteristic(ROC) curve and area under the curve(AUC) were conducted to evaluate the diagnostic value of SNHG1 in cervical cancer.Result The relative expression of SNHG1 mRNA in the peripheral blood of cervical cancer patients was significantly higher than that in the control group,and the relative expression of SNHG1 mRNA in cervical cancer tissues was significantly higher than that in adjacent tissues,and the differences were statistically significant(P<0.01).The results of ROC curve indicated that the AUC of SNHG1 expression for diagnosis of cervical cancer was 0.836(95%CI:0.748-0.924),and the difference was statistically significant(P<0.01).The relative expression level of SNHG1 mRNA in peripheral blood was 0.551 as the boundary.There were 30 patients with cervical cancer with high SNHG1 expression and 26 patients with cervical cancer with low SNHG1 expression.Cervical cancer patients with clinical stages Ⅰ-Ⅱ,without lymph node metastasis or recurrence after surgery showed low expression of SNHG1 in penpheral blood,and the difference was statistically significant(P<0.05).The results of Spearman correlation analysis showed that the expression of SNHG1 in the peripheral blood of cervical cancer patients was positively correlated with the expression of SNHG1 in cervical cancer tissues(r=0.785,P<0.01).The overall survival of cervical cancer patients with high SNHG1 expression was significantly shorter than that of patients with low SNHG1 expression,and the difference was statistically significant(P<0.01).Conclusion SNHG1 expression was upregulated in peripheral blood cells from cervical cancer patients,and it could be listed as an important and potential biomarker for diagnosis and prognosis of this lethal disease.
