直肠癌组织中单羧酸转运蛋白1、单羧酸转运蛋白4的表达情况及与患者临床特征和预后的关系

【摘要】 目的探讨直肠癌组织中单羧酸转运蛋白1(MCT1)、单羧酸转运蛋白4(MCT4)的表达情况及与患者临床特征和预后的关系。方法采用免疫组织化学染色法检测106例直肠癌患者直肠癌组织中MCT1、MCT4的表达情况,分析其与患者临床特征及预后的关系。对Ⅲ期直肠癌患者(术后均接受辅助性放化疗)进行亚组分析,比较不同MCT1、MCT4表达情况的Ⅲ期直肠癌患者的生存预后。结果 106例患者中,57例患者MCT1呈高表达,48例患者MCT4呈高表达。不同临床分期、淋巴结转移情况、血管浸润情况、术后5年内复发和(或)转移情况的直肠癌患者直肠癌组织中MCT1和MCT4的表达情况比较,差异均有统计学意义(P﹤0.05)。MCT1低表达患者的生存情况优于MCT1高表达患者,MCT4低表达患者的生存情况优于MCT4高表达患者,差异均有统计学意义(P﹤0.05)。MCT1低表达的Ⅲ期直肠癌患者的生存情况优于MCT1高表达的Ⅲ期直肠癌患者,MCT4低表达的Ⅲ期直肠癌患者的生存情况优于MCT4高表达的Ⅲ期直肠癌患者,差异均有统计学意义(P﹤0.05)。结论直肠癌组织中MCT1和MCT4表达情况与患者临床分期、淋巴结转移情况、血管浸润情况及生存情况有关,可将MCT1、MCT4作为直肠癌预后预测的重要指标。

【Abstract】 Objective To explore the expression of monocarboxylate transporter 1(MCT1), monocarboxylate transporter 4(MCT4) in rectal cancer tissues and their relationship with patients’ clinical characteristics and prognosis.Method Immunohistochemical staining was performed to detect the expression of MCT1 and MCT4 in rectal cancer tissues from 106 cases with rectal cancer, and the relationship with clinical characteristics and prognosis was assessed. A subgroup(receiving postoperative adjuvant chemoradiotherapy) analysis involving patients with stage III diseases was conducted in order to evaluate the prognosis among those with stage III lesions and varying expression of MCT1 and MCT4. Result Of the 106 patients, 57 cases had high expression of MCT1 and 48 cases were with high expression of MCT4. Clinical stage, lymph node metastasis, vascular infiltration, recurrence and/or metastasis within 5 years after surgery were associated with the MCT1 and MCT4 expression in rectal cancer patients(P<0.05). Low MCT1 expression was related to better survival as compared with high MCT1 expression, likewise, lower MCT4 demonstrated better survival than high MCT4 expression(P<0.05). Furthermore, in patients with stage III lesions, improved survival was observed in those with low MCT1 expression versus high expression, similar findings were noted for low MCT4 versus high MCT4 expression in the subgroup of stage III diseases(P<0.05). Conclusion The expression of MCT1 and MCT4 are correlated with clinical stage, lymph node metastasis, vascular infiltration, and survival, suggesting that their predictive value in evaluating the prognosis of rectal cancer.
