【摘要】 目的探讨微小RNA(miRNA)-214在胃癌组织中的表达情况及临床意义。方法选取47例胃癌患者的胃癌组织标本及对应的癌旁组织标本,采用实时荧光定量聚合酶链反应(qRT-PCR)检测胃癌组织(n=47)和癌旁组织(n=47)中miRNA-214的表达情况,分析miRNA-214表达与胃癌患者临床特征的关系。结果胃癌患者胃癌组织中miRNA-214的相对表达量明显高于癌旁组织(P <0.01)。不同TNM分期、分化程度、淋巴结转移情况的胃癌患者胃癌组织中miRNA-214的相对表达量比较,差异均有统计学意义(P <0.05);不同性别、年龄、肿瘤直径的胃癌患者胃癌组织中miRN A-214的相对表达量比较,差异均无统计学意义(P> 0.0 5)。3年无瘤生存的23例胃癌患者胃癌组织中miRNA-214的相对表达量与3年复发或转移的24例胃癌患者胃癌组织中miRNA-214相对表达量比较,差异有统计学意义(P <0.05)。miRNA-214高表达、TNM分期为Ⅲ～Ⅳ期、有淋巴结转移和低分化均是胃癌患者3年无瘤生存的独立危险因素(P <0.05)。结论 miRNA-214在胃癌组织中呈相对高表达,miRNA-214高表达、TNM分期为Ⅲ～Ⅳ期、有淋巴结转移和低分化均是胃癌患者预后的独立危险因素。更多还原

【Abstract】 Objective To investigate the expression and clinical significance of microRNA-214(miRNA-214) in gastric cancer.Method The specimens of gastric cancer tissues and adjacent tissues of 47 patients with gastric cancer were selected.The quantitative real-time polymerase chain reaction(qRT-PCR) was utilized in detecting the expression of miRNA-214 in gastric cancer tissues(n=47) and adjacent tissues(n=47),besides,the relationship between the expression of miRNA-214 and the clinical characteristics of gastric cancer patients was determined.Result The relative expression of miRNA-214 in gastric cancer tissues was significantly higher than that in normal tissues(P<0.01).The expression of miRNA-214 varied among patients with different TNM stage,degree of differentiation and lymph node metastasis(P<0.05);while it was independent of sex,age and tumor diameter in patients with gastric cancer(P>0.05).The relative expression level of miRNA-214 in gastric cancer tissues of 23 patients with 3-year tumor-free survival was statistically significant compared with that of 24 patients with 3-year recurrence or metastasis(P<0.05).High expression of miRNA-214,TNM stage of Ⅲ-Ⅳ,and positive lymph node metastasis as well as lower degree of differentiation are the independent risk factors for the 3-year tumor-free survival of the patients with gastric cancer(P<0.05).Conclusion The relative high expression of miRNA-214 is present in the gastric cancer tissues,besides,high expression of miRNA-214,TNM stage ofⅢ-Ⅳ,and positive lymph node metastasis as well as lower degree of differentiation are the independent risk factors for the prognosis of the patients with gastric cancer.更多还原
