绵羊微卫星标记多重PCR体系的建立与应用
Establishment and Application of Multiplex-PCR System for Sheep Microsatellite Markers
【摘要】 微卫星标记由于具有在基因组中分布广、多态性高及共显性等特点,在群体遗传多样性分析中广泛应用。本试验针对绵羊19个微卫星标记位点构建了8个多重PCR扩增体系,其中包括3个三重,5个两重PCR扩增体系。采用建立的多重PCR扩增体系对32只小尾寒羊的多态性进行了检测。结果表明,19个微卫星标记的等位基因数在5~17之间,期望杂合度、观测杂合度和多态信息含量分别位于0.423~0.885、0.344~0.875和0.392~0.859之间。本试验所建立的多重PCR扩增体系将为绵羊遗传多样性、亲子鉴定和个别识别提供技术基础。
【Abstract】 Microsatellite markers have been widely used in population genetic diversity analysis due to their wide distribution,high polymorphism and co-dominance. In this study, eight multiplex-PCR systems were established for 19 microsatellite markers in sheep, including three triplex-PCR and five duplex-PCR systems. Further, genetic polymorphism was examined in 32 smalltailed han sheep by multiplex-PCR system developed in this study. The results showed that the number of alleles of 19 microsatellite markers ranged from 5 to 17. The expected heterozygosity, observed heterozygosity and polymorphism information content were 0.423-0.885, 0.344-0.875 and 0.392-0.859, respectively. The multiplex PCR amplification system established in this experiment will provide technical basis for sheep genetic diversity, paternity identification and individual identification.
