云南不同来源四路糯玉米waxy和tb1基因分析
The Waxy and tb1 Gene Analysis of Four-Row Wax Maizes with Different Sources of Yunnan
【摘要】 [目的]为四路糯资源的保护、研究与开发利用特异资源提供科学依据。[方法]对8份云南不同来源的四路糯资源材料进行糯性基因waxy、分蘖基因tb1的测序及SSR分子标记分析,探讨不同来源四路糯的形成规律。[结果]试验所用糯玉米具有共同的突变类型wx-D10,即waxy基因第10外显子15-bp的缺失;检测控制分蘖的tb1基因发现,在四路糯中保留了2类不同的等位基因,并发现具有杂合基因型的植株存在;进一步利用40对SSR标记检测不同来源的四路糯,获得了27个多态性基因位点。[结论]试验所用四路糯有统一的起源,在形成后基因组内依然保留了大量多等位基因位点,由于四路糯在传播过程中发生不同等位基因的丢失,形成不同类型的四路糯。
【Abstract】 [Objective]To provide scientific basis for special germplasm resources of the protection,development and utilization of four-row waxy maize resources.[Method]Sequencing of waxy gene waxy,tillering gene tb1 and SSR molecular marker analysis were carried out for eight four-row wax maize resources of Yunnan.Their formation rules were also discussed.[Result]These four-row waxy maizes had the same mutation type wx-D10,which was 15-bp deletion in exon 10 of waxy gene; detection of tb1 gene controlling tillering revealed that two different alleles were retained in four-row waxymaizes and plants with heterozygous genotype were found; 40 SSR markers were used to detect four-row waxy maizes from different sources,and 27 polymorphic loci were obtained.[Conclusion]These four-row waxy maizes had a unified origin; many different alleles were still retained in the genome after the formation of four-row waxy maizes; different types of four-way waxy were formed due to the loss of alleles during the process of material transmission.
