利用mtCOI PCR-RFLP技术鉴别草地贪夜蛾水稻型和玉米型
Rapid Differentiation of Corn- and Rice-Strain of Spodoptera furgiperda using mtCOI PCR-RFLP Technique
【摘要】 本研究基于mtCOI基因,利用相关软件及系统发育分析,筛选鉴别水稻型和玉米型草地贪夜蛾的酶切位点,并利用PCR-RFLP技术对两者进行鉴定。结果表明:水稻型个体的mtCOI基因序列在154~159 bp处存在SacⅠ酶切位点,玉米型则无该酶切位点;利用mtCOI PCR-RFLP技术可以有效鉴别草地贪夜蛾的水稻型和玉米型。本研究结果可为草地贪夜蛾水稻型、玉米型的快速鉴别及其精准防控提供参考。
【Abstract】 The restriction enzyme cutting site identifying rice-and corn-strain individuals of Spodoptera frugiperda was screened out by related software and phylogenetic analyses based on mtCOI sequences, and was used to differentiate the two strains. The results showed that the mtCOI sequence of rice-strain individual existed restriction enzyme cutting site of Sac Ⅰ at 154~159 bp, but corn-strain individual did not have the above site. The rice-and corn-strain individuals of S. frugiperda could be effectively differentiated by mtCOI PCR-RFLP technology. The results could provide references for the rapid identification and precision control of S. frugiperda.
