柴胡-白芍异病同治分子机制的网络药理学分析

【摘要】 目的应用网络药理学方法筛选柴胡-白芍药对治疗多种疾病的活性成分、作用靶点及通路,探讨其异病同治的分子机制。方法应用中药系统药理学数据库(TCMSP)和UniProt数据库筛选出柴胡-白芍药对的活性成分及对应靶点;采用Cytoscape 3.6.0软件构建柴胡-白芍药对的活性成分-靶点网络;运用STRING平台构建靶蛋白相互作用(PPI)网络;借助生物学信息注释数据库(DAVID)对靶点进行GO生物过程富集和KEGG信号通路分析。结果柴胡-白芍药对中20种活性成分作用于193个潜在靶点;参与正向调节转录因子的活性、负调控细胞生物学过程、细胞复合蛋白介导的蛋白质复合物组装等7个主要GO生物过程;涉及免疫、细胞信号转导、癌症、神经、传染病等67条信号通路。结论预测了柴胡-白芍药对在胃肠炎症、神经系统、癌症等多种疾病上的治疗作用及其药效物质基础和分子机制,阐释了柴胡-白芍药对异病同治的分子基础。

【Abstract】 Objective The active ingredients,targets and pathways of Bupleuri radix combined with Paeoniae alba radix in the treatment of various diseases were screened by network pharmacological method, and the molecular mechanism of "the same treatment for different diseases" was discussed. Methods TCMSP and UniProt databases were used to screen the active ingredients and corresponding targets of Bupleuri radix-Paeoniae alba radix herb pair.Cytoscape 3.6.0 software was used to construct the active ingredient-target network. Target protein interaction network was constructed on STRING platform. GO biological process enrichment and KEGG signal pathway analysis were carried out on the targets with the help of biological information annotation database(DAVID). Results The 20 active ingredients of Bupleuri radix-Paeoniae alba radix herb pair act on 193 potential targets. They involved in seven major GO biological processes included positive regulation of transcription factor activity,negative regulation of cellular process,cellular chaperone-mediated protein complex assembly. These targets also involved 67 signaling pathways such as immune,signal transduction,cancer,nerves,and infectious diseases. Conclusion Our study predicted the therapeutic value of Bupleuri radix-Paeoniae alba radix herb pair on gastrointestinal inflammation,nervous system, cancer and other diseases, as well as its pharmacodynamic material basis and molecular mechanism, and clarified the molecular basis of Bupleuri radix-Paeoniae alba radix herb pair on "the same treatment for different diseases".
