HPLC法同时测定小儿消积化虫散中6种成分的含量
Simultaneous Determination of 6 Components in Xiao’er Xiaoji Huachong Powder by HPLC
【摘要】 目的:建立HPLC梯度洗脱法同时测定小儿消积化虫散中天名精内酯酮、莪术二酮、莪术醇、去氢土莫酸、猪苓酸C和茯苓酸的含量。方法:采用Agilent Eclipse XDB-C18色谱柱(250 mm×4. 6 mm,5μm),以乙腈-0. 05%磷酸溶液为流动相,梯度洗脱,检测波长为210 nm,流速为0. 9 ml·min-1,柱温为30℃,进样体积为10μl。结果:天名精内酯酮、莪术二酮、莪术醇、去氢土莫酸、猪苓酸C和茯苓酸分别在1. 19~29. 75,2. 46~61. 50,1. 52~38. 00,3. 09~77. 25,0. 87~21. 75,3. 78~94. 50μg·ml-1范围内线性关系良好,r均≥0. 999 2;平均加样回收率分别为97. 76%,98. 07%,98. 70%,99. 49%,96. 98%和100. 07%,RSD分别为1. 31%,1. 06%,1. 26%,0. 75%,1. 41%和0. 62%(n=6)。结论:本文所建立的方法操作简便、数据准确、重复性好,能够同时对小儿消积化虫散中6种成分进行含量测定,为提高和完善小儿消积化虫散的质量控制标准提供了有效的方法。
【Abstract】 Objective: To establish an HPLC gradient elution method for the simultaneous determination of carabrone,curdione,curcumol,dehydrotumulosic acid,polyporenic acid C and pachymic acid in Xiao’er Xiaoji Huachong powder. Methods: The separation was carried out on an Agilent Eclipse XDB-C18 chromatographic column (250 mm × 4. 6 mm,5 μm). The mobile phase consisted of acetonitrile-0. 05% phosphoric acid solution with gradient elution. The detection wavelength was set at 210 nm. The flow rate was0. 9 ml·min-1,the temperature of column was set at 30 ℃,and the injection volume was 10 μl. Results: The linear range of carabrone,curdione,curcumol,dehydrotumulosic acid,polyporenic acid C and pachymic acid was 1. 19-29. 75,2. 46-61. 50,1. 52-38. 00,3. 09-77. 25,0. 87-21. 75 and 3. 78-94. 50 μg · ml-1 (r≥0. 999 2),respectively. The average recovery was 97. 76%,98. 07%,98. 70%,99. 49%,96. 98% and 100. 07%,and the RSD was 1. 31%,1. 06%,1. 26%,0. 75%,1. 41% and 0. 62% (n =6),respectively. Conclusion: The method with good reproducibility is simple and accurate,which can be applied in the simultaneous determination of six components in Xiao’er Xiaoji Huachong powder,and supplies another effective method for improving and perfecting the quality control of Xiao’er Xiaoji Huachong powder.
