中药逐水饮对小鼠Lewis肺癌胸腔积液模型积液生成及Th17免疫平衡的影响
Effects of Chinese Medicine Zhushui Decoction on Hydrothorax Formation and Th17 Immune Balance of Lewis Lung Cancer Pleural Effusion Model in Mice
【摘要】 目的观察中药逐水饮对小鼠Lewis肺癌胸腔积液模型积液生成及Th17细胞免疫平衡的影响。方法小鼠随机分为正常对照组、模型组、逐水饮组、顺铂组、逐水饮+顺铂组。采用胸腔注射Lewis肺癌单细胞悬液的方法建立小鼠Lewis肺癌胸腔积液模型。给药结束处死小鼠,记录模型小鼠恶性胸腔积液(MPE)体积、胸壁转移瘤计数、转移瘤质量,采用流式细胞术法检测MPE和外周血中Th17、Treg细胞百分比,ELISA法检测MPE和外周血中白细胞介素17(IL-17)、白细胞介素-23(IL-23)、转化生长因子β1(TGF-β1)表达水平。结果与模型组比较,逐水饮组、顺铂组和逐水饮+顺铂组小鼠MPE体积、胸壁转移瘤数目减少(P <0.05),转移瘤质量减轻(P <0.05),生存期延长(P <0.05),其中逐水饮+顺铂组变化更加明显(P <0.05)。与正常对照组比较,模型组外周血中Th17细胞百分比及IL-17、IL-23水平降低(P <0.05),Treg细胞百分比及TGF-β1水平升高(P <0.05);与模型组比较,逐水饮组、顺铂组和逐水饮+顺铂组小鼠MPE和外周血中Th17细胞百分比及IL-17、IL-23水平升高(P <0.05),MPE中Treg细胞百分比及TGF-β1水平降低(P <0.05);逐水饮+顺铂组小鼠MPE和外周血中Th17细胞百分比及IL-17/IL-23水平高于逐水饮组,Treg细胞百分比及TGF-β1水平低于逐水饮组,但差异无统计学意义(P> 0.05);模型小鼠MPE中Th17、Treg细胞百分比均高于其外周血中Th17、Treg细胞百分比(P <0.05)。结论中药逐水饮能够抑制小鼠MPE生成,纠正Th17细胞失衡,减轻MPE微环境免疫抑制状态,增强小鼠对肿瘤的免疫作用。
【Abstract】 Objective:To observe the effects of Chinese medicine Zhushui Decoction on the formation of pleural effusion and Th17 cell immune balance in pleural effusion model of Lewis lung cancer in mice. Methods:The mice were randomly divided into the normal control group,the model group,Zhushui Decoction group,cisplatin group,Zhushui Decoction group+cisplatin group. The pleural effusion model of Lewis lung cancer in mice was established by intrathoracic injection of Lewis lung cancer single cell suspension. The mice were killed at the end of administration;the volume of malignant pleural effusion(MPE),the number and the weight of metastatic tumors in the chest wall were recorded;the percentages of Th17 and Treg cells in MPE and peripheral blood were detected by flow cytometry;the expression levels of IL-17,IL-23 and TGF-β1 in MPE and peripheral blood were detected by ELISA. Results:The results of chest CT scan and dissection showed that the lung field was clear and the pleura was smooth in the normal control group,while pleural effusion was formed in the model group;compared with the model group;the MPE volume of mice and the number of chest wall metastases in Zhushui Decoction group,cisplatin group and Zhushui Decoction+cisplatin group were decreased(P < 0.05);the weight of metastases decreased(P < 0.05),and the survival time prolonged(P < 0.05),especially in the Zhushui Decoction+cisplatin group(P < 0.05);compared with the normal control group,the percentage of Th17 cells and the levels of IL-17 and IL-23 in peripheral blood of the model group decreased(P < 0.05),while the percentage of Treg cells and the level of TGF-β1 increased(P < 0.05);compared with the model group,the percentage of Th17 cells and the levels of IL-17 and IL-23 in MPE and peripheral blood of mice in the Zhushui Decoction group,cisplatin group and Zhushui Decoction+cisplatin group increased(P < 0.05),while the percentage of Treg cells and the level of TGF-β1 in MPE decreased(P < 0.05);the percentage of Th17 cells and the levels of IL-17 and IL-23 in MPE and peripheral blood of mice in the Zhushui Decoction + cisplatin group were higher than those in the Zhushui Decoction group,while the percentage of Treg cells and the level of TGF-β1 were lower than those in the Zhushui Decoction group,but there was no significant difference(P > 0.05);the percentages of Th17 and Treg cells in MPE of model mice was higher than those in peripheral blood(P<0.05). Conclusion:Chinese medicine Zhushui Decoction can inhibit the production of MPE in mice,correct the imbalance of Th17 cells,alleviate the immunosuppression of MPE microenvironment,and enhance the immune effect on tumors.
