参葵通脉颗粒对慢性心衰大鼠模型心肌细胞焦亡及相关因子的影响

Effects of Shenkui Tongmai Granule on the Charred Death of Myocardial Cells and Related Factors in Rats with Chronic Heart Failure

【摘要】 目的:探讨参葵通脉颗粒治疗慢性心衰(CHF)的作用机制。方法:腹主动脉缩窄制备CHF模型,随机分为模型组、参葵通脉颗粒低、中、高剂量组、贝那普利组,另以10只不缩窄腹主动脉大鼠为假手术组,根据分组给予相应药物,每日1次共4周。彩色超声系统检测LVEF与FS,化学发光法检测NT-proBNP,TUNEL法检测心肌程序性坏死程度,western blot检测TLR4、NF-κB、Caspase 1及IL-1β。结果:与假手术组比较,模型组LVEF、FS明显下降,NT-proBNP、TUNEL阳性细胞、TLR4、NF-κB、Caspase 1、IL-1β表达明显增加,与模型组比较,参葵通脉颗粒各组LVEF、FS明显上升,NT-proBNP、TUNEL阳性细胞、TLR4、NF-κB、Caspase 1、IL-1β表达明显降低。结论:参葵通脉颗粒可改善CHF大鼠模型心脏功能,降低心肌细胞焦亡与TLR4、NF-κB、Caspase 1、IL-1β的表达。

【Abstract】 Objective:To explore the possible mechanism of Shenkui Tongmai Granule in the treatment of chronic heart failure.Methods:Rats with chronic heart failure were prepared by abdominal aorta coarctation.They were randomly divided into model group,Shenkui Tongmai low dose group,middle dose group,high dose group and positive drug group.The other 10 rats without abdominal aortic stenosis were served as sham operation group.The drugs were given according to the group,one time a day,and all the groups were intervened for 4 weeks.LVEF and FS were detected by color ultrasound system.NT-pro BNP was detected by chemiluminescence method.Programmed myocardial necrosis was detected by TUNEL staining.TLR4,NF-kappa B,Caspase 1 and IL-1 beta were detected by Western blot.Results:Compared with the normal control group,LVEF and FS in the model group decreased significantly;NT-proBNP,TUNEL-positive cells,the expression of TLR4,NF-kappa B,Caspase 1,IL-1 beta increased significantly.Compared with the model group,LVEF and FS in the Shenkui Tongmai granule groups increased significantly;NT-proBNP,TUNEL-positive cells,the expression of TLR4,NF-kappa B,Caspase 1,IL-1 beta decreased significantly.Conclusion:Shenkui Tongmai Granule can improve the cardiac function of CHF rat model and reduce the pyroptosis of cardiomyocytes and the expression of TLR4,NF-kappa B,Caspase 1 and IL-1 beta.
