脐带间充质干细胞移植对大鼠脊髓损伤区域神经损伤的修复效果分析
Repair Effect of Umbilical Cord Mesenchymal Stem Cells Transplantation on Nerve Injury in Rats with Spinal Cord Injury
【摘要】 目的:探究脐带间充质干细胞(UMSCs)移植对大鼠脊髓损伤区域神经损伤的修复效果。方法:选择70只大鼠,雄性,体重220~260 g,采用随机数字法分为研究组和对照组,各35只。两组大鼠均采用脊髓半切建模。建模1周后研究组行UMSCs移植,对照组给予等量不含干细胞0.9%氯化钠溶液注射。移植24 h及1、2、4周后,比较两组Tarlov评分。移植2周后,观察两组的组织学情况,比较两组血清白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果:移植24 h后两组Tarlov评分均较低,差异无统计学意义(P>0.05),移植1、2、4周后研究组Tarlov评分均明显高于对照组(P<0.05)。移植2周后,对照组大鼠脊髓组织学检查发现,脊髓空洞面积大,神经纤维相对稀疏,存在不同程度的炎性细胞浸润且星型胶质细胞明显聚集增生;研究组炎性细胞浸润且星型胶质细胞与对照组比较明显降低,且有神经纤维通过大鼠脊髓损伤区域。研究组移植2周后两组IL-1比较,差异无统计学意义(P>0.05),研究组IL-6和TNF-α水平均明显低于对照组(P<0.05)。结论:采用UMSCs在一定程度上可以减轻大鼠损伤脊髓部位的炎性细胞浸润、抑制胶质细胞形成瘢痕组织,对于改善大鼠神经损伤后功能的恢复有着重要意义。
【Abstract】 Objective: To investigate the repair effect of umbilical cord mesenchymal stem cells(UMSCs) transplantation on nerve injury in rats with spinal cord injury. Method: A total of 70 rats were selected, males weight was 220-260 g. They were divided into study group and control group according to the random number table method, 35 rats in each group. Both groups were modeled by spinal cord semisection. One week after modeling, the study group was given UMSCs transplantation and the control group was given equal amount of 0.9% sodium chloride solution injection without stem cells. The Tarlov scores were compared between the two groups 24 h, 1, 2 and 4 weeks after transplantation. Two weeks after transplantation, the histology of the two groups was observed and the serum levels of interleukin-1(IL-1), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were compared. Result: Tarlov score was lower in both groups 24 h after transplantation, the difference was not statistically significant(P>0.05). The Tarlov score of the study group was significantly higher than those of the control group after 1, 2 and 4 weeks of transplantation(P<0.05). Two weeks after transplantation, histological examination of the spinal cord in the control group showed that the area of the spinal cavity was large, the nerve fibers were relatively sparse, there were inflammatory cell infiltration of different degrees, and astrocytes were obviously clustered and proliferated. In the study group, inflammatory cell infiltration and astrocytes were significantly lower than those in the control group, and nerve fibers passed through the spinal cord injury area of the rats. There was no significant difference in IL-1 between the two groups 2 weeks after transplantation(P>0.05). And IL-6 and TNF-α levels in the study group were significantly lower than those in the control group(P<0.05).Conclusion: UMSCs can alleviate the inflammatory cell infiltration at the site of spinal cord injury in rats and inhibit the formation of scar tissue in glial cells, which is important to improve the recovery of function after nerve injury in rats.
