酸枣仁中不同单体组合对小鼠睡眠的影响及其机制
Effects of monomer combinations in semen zizyphi spinosae on sleep in mice and their mechanisms
【摘要】 目的:研究酸枣仁中不同单体斯皮诺素(S)、酸枣仁皂苷A(A)、酸枣仁皂苷B(B)组合对小鼠的镇静催眠作用是否存在协同效果。方法:采用自主活动实验检测SA,SB,AB和SAB组对小鼠自主活动的影响;通过使用睡眠解析软件SleepSign完成对EEG和EMG数据的采集记录,对小鼠的睡眠觉醒状态进行判定;通过c-Fos免疫组化探讨作用机制。结果:SA,SB,AB和SAB组能减少小鼠自主活动,并与生理盐水组比较有显著性差异; AB组和SA组不改变小鼠睡眠量。SB组和SAB组能增加小鼠NREM睡眠量和减少觉醒量,与AB组、SA组和生理盐水组有显著性差异; SB组和SAB组能激活VLPO睡眠中心,并且抑制VTM觉醒中心。结论:有效单体组合SB和SAB对睡眠有很好的促进作用,其机制可能与GABA有关,并推导出B和S可能有协同增强作用。
【Abstract】 Objective: To study the synergistic effect of different combinations of monomers spinosin(S),jujuboside A(A),and jujuboside B(B) in semen zizyphi spinosae on sedation and hypnosis of mice. Methods:The effects of SA,SB,AB,and SAB combinations on autonomous activity of mice were detected by autonomous activity test,and the sleep awakening state of the recorded mice was determined by using SleepSign software,which was used to collect and record the data of EEG and EMG. The mechanism of affect was discussed by c-Fos immunohistochemistry. Results: The SA,SB,AB,and SAB combinations could reduce the autonomous activity of mice,and there were significant differences between the 4 groups and the normal saline group. The sleep time in the AB group and SA group did not change. The SB and SAB combinations could significantly increase NREM sleep and decrease arousal in the mice,compared with AB,SA and normal saline groups. The SB and SAB combinations could activate the VLPO sleep center and inhibit the VTM arousal center. Conclusion: The SB and SAB combinations can promote sleep,and their mechanisms may be related to GABA,and it is deduced that B and S may have synergistic enhancement effect.
