沙棘黄酮在大鼠子宫缺血再灌注损伤中的作用研究

【摘要】 目的研究沙棘黄酮在大鼠子宫抗缺血再灌注损伤中的作用。方法 48只清洁级Wistar大鼠,随机分为A组、B组、C组、D组,每组12例。线栓法制备缺血再灌注损伤模型, B组于术前10 d起给予腹腔注射丹参酮ⅡA注射液20 mg/(kg·d);C组于术前10 d起给予0.3 g/kg沙棘黄酮稀释于3 ml生理盐水灌胃;D组于术前10 d起给予0.6 g/kg沙棘黄酮稀释于3 ml生理盐水灌胃;A组给予等量生理盐水灌胃。缺血再灌注2 h取大鼠子宫组织,利用分光光度计测量大鼠子宫组织匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)水平,免疫组化染色法(SABC)检测Bax、Bcl-2、caspase-3蛋白的表达。比较四组大鼠子宫组织MDA、SOD水平及Bax、Bcl-2和caspase-3蛋白表达水平。结果 B组、C组、D组大鼠子宫组织MDA水平分别为(69.8±8.9)、(66.5±7.9)、(63.6±7.2)μmol/L,低于A组的(110.6±13.8)μmol/L;SOD水平分别为(362.4±21.6)、(376.1±23.5)、(380.8±24.4)μmol/L,高于A组的(100.8±9.6)mkat/L,差异具有统计学意义(P<0.05);B组、C组、D组大鼠子宫组织MDA、SOD水平比较差异无统计学意义(P>0.05)。B组、C组、D组大鼠子宫组织Bax、caspase-3蛋白表达均低于A组, Bcl-2蛋白表达水平高于A组,差异具有统计学意义(P<0.05);B组、C组、D组Bax、Bcl-2和caspase-3蛋白表达水平比较差异无统计学意义(P>0.05)。结论沙棘黄酮对大鼠子宫缺血再灌注损伤具有保护作用,沙棘黄酮的剂量差异对子宫缺血再灌注损伤的保护作用无差异。

【Abstract】 Objective To study the effect of seabuckthorn flavone on uterine ischemia-reperfusion injury in rats. Methods A total of 48 clean-grade Wistar rats were randomly divided into group A, group B, group C and group D, with 12 rats in each group. Ischemia-reperfusion injury model was prepared by thread embolization. Group B was given intraperitoneal injection of tanshinone Ⅱ A 20 mg/(kg·d) at 10 d before operation, group C was given gavage administration of 0.3 g/kg seabuckthorn flavonoids diluted in 3 ml saline at 10 d before operation, group D was given gavage administration of 0.6 g/kg seabuckthorn flavonoids diluted in 3 ml saline at 10 d before operation, and group A was given gavage administration of the same amount of saline. After 2 h of ischemia-reperfusion, the uterus tissue of rats was taken and the levels of malondialdehyde(MDA) and superoxide dismutase(SOD) in uterus homogenate were measured by spectrophotometer, and the expression of Bax, Bcl-2 and caspase-3 proteins was detected by immunohistochemical staining(SABC). The levels of MDA, SOD and the expression of Bax, Bcl-2 and caspase-3 in uterus of rats in four groups were compared. Results The MDA level in uterine tissue of rats were(69.8±8.9),(66.5±7.9) and(63.6±7.2) μmol/L respectively in group B, group C and group D, which was lower than(110.6±13.8) in group A; SOD level were(362.4±21.6),(376.1±23.5) and(380.8±24.4) mkat/L respectively, which was higher than(100.8±9.6) mkat/L in group A. Their difference was statistically significant(P<0.05). There was no statistically significant difference in MDA and SOD level in uterine tissue of rats of group B, group C and group D(P>0.05). The expression of Bax and caspase-3 protein in uterine tissue of rats of group B, group C and group D was lower than that in group A, and expression of Bcl-2 protein was higher than that in group A. Their difference was statistically significant(P<0.05). There was no statistically significant difference in expression of Bax, Bcl-2 and caspase-3 protein of group B, group C and group D(P>0.05). Conclusion Seabuckthorn flavone has protective effects on uterine ischemia-reperfusion injury in rats. There is no difference in the protective effects of different doses of seabuckthorn flavone on uterine ischemia-reperfusion injury.
