内侧隔核胆碱能神经元参与SNI神经病理性疼痛大鼠模式分离障碍
PARTICIPATION OF CHOLINERGIC NEURONS IN MEDIAL SEPTUM IN PATTERN SEPARATION DISORDER IN NEUROPATHIC PAIN RATS WITH SPINAL NERVE INJURY
【摘要】 目的:研究部分神经损伤(spared nerve injury, SNI)神经病理性疼痛大鼠导致模式分离(pattern separation)障碍的机制。方法:实验分为两部分,第一部分实验大鼠分为两组:假手术(sham SNI)组、SNI组;第二部分实验大鼠分为两组:假损毁、SAP损毁。首先制作SNI神经病理性疼痛模型,之后用八臂迷宫进行模式分离实验来比较sham SNI和SNI实验组大鼠的空间认知水平。此外用SAP (192Ig G-saporin)的方式损毁内侧隔核(medial septum, MS)内的胆碱能神经元,用八臂迷宫来比较损毁组和对照组大鼠的模式分离变化。结果:SNI组大鼠在八臂迷宫中的正确率明显低于sham SNI组,并且SNI导致内侧隔核内的胆碱能神经元数目降低,而损毁MS内胆碱能神经元后大鼠八臂迷宫的正确率明显低于假损毁组。结论:SNI神经病理性疼痛导致模式分离障碍可能与内侧隔核中的胆碱能神经元有关。
【Abstract】 Objective: To study the mechanism of pattern separation disorder in rats with neuropathic pain caused by spinal nerve injury(SNI). Methods: The experiments were divided into two parts. Rats in the first part were divided into two groups: sham SNI and SNI, and rats in the second part were divided into two groups: sham lesion and SAP lesion. Neuropathic pain model was established with spinal nerve injury and cholinergic neurons in the medial septum(MS) were lesioned by 192 IgG-saporin(SAP). The spatial cognitive impairment was compared by pattern separation experiment using eightarm maze. Results: The percent of correct choice in the eight-arm maze in the SNI group was significantly lower than that in the sham SNI group, and SNI-induced neuropathic pain reduced the number of cholinergic neurons in medial septum. While the percent of correct choice was significantly lower in the SAP lesioned group than that in the sham lesion group. Conclusion: The disturbance of pattern separation in neuropathic pain rats with SNI maybe related to cholinergic neurons in medial septum.
