【摘要】 目的检测特应性皮炎(AD)患者外周血中辅助性T细胞22(Th22)和调节性T细胞(Treg)及其细胞因子IL-22和TGF-β的表达,探讨其与AD发病之间的关系及临床意义。方法选择52例AD患者,按照欧洲特应性皮炎评分标准(SCORAD)进行评分分组;使用流式细胞仪检测52例AD患者和和20例健康人Th22和Treg细胞的数量;用酶联免疫吸附法(ELISA)检测血清中IL-22和TGF-β的表达。结果与对照组比较,AD患者外周血中Th22细胞的比例及血清中IL-22的水平显著增高(P<0.0001),Treg细胞的比例及血清中TGF-β的水平显著降低(P<0.0001);与中度AD患者比较,重度AD患者外周血Th22细胞的比例及血清中IL-22的水平显著增高(P<0.0001),Treg细胞的比例及血清中TGF-β的水平显著降低(P<0.0001)。结论 AD患者外周血中Th22细胞及其相关细胞因子表达水平显著升高,而Treg细胞及其相关细胞因子的表达降低,提示Th22和Treg可能在AD的发病机制中发挥重要作用。更多还原

【Abstract】 Objective To detect the variations of T helper 22 cells(Th22)and regulatory T cells(Treg)and its related cytokines interleukin(IL)-22 and transforming growth factor(TGF-β)in peripheral blood of the patients with atopic dermatitis(AD).Methods 52 patients with AD were recruited and grouped according to the European atopic dermatitis criteria rating(SCORAD).The percentages of Th22 and Treg cells in 52 AD patients and 20 control group was detected by flow cytometric.The concentrations of IL-22 and TGF-βin serum was detected by enzyme-linked immunosorbent assay(ELISA).Results The proportion of Th22 cells in peripheral blood and the level of IL-22 in serum of patients with AD weresignificantly higher than normal people(P<0.05,P<0.0001).The proportion of Th22 cells and the level of IL-22 in severe AD patients were higher than those in moderate AD patients(P<0.05,P<0.0001).In addition,the proportion of Treg cells in peripheral blood and the level of TGF-βserum of patients with AD were significantly lower than those in normal patients(P<0.05,P<0.0001).The proportion of Treg cells and the level of TGF-βin patients with severe AD were significantly lower than those in moderate AD patients(P<0.05,P<0.0001).Conclusion The study suggests that Th22 and Treg cell might play an important role in the pathogenesis of AD.更多还原
