COMPARATIVE ANALYSIS OF JAPANESE ENCEPHALITIS VIRUS INFECTION IN MACROPHAGES WITH DIFFERENT POLARIZED PHENOTYPES
【Abstract】 The effect of macrophages with different activated phenotypes on Japanese encephalitis virus(JEV) infection was studied with titration(TCID50), immunofluorescence assay and real-time qPCR. Mouse macrophages were polarized by different cytokines including IFN-γ or IL-4 and the role of NO production during JEV infection was analyzed in terms of expression of iNOS and Arg1 by using realtime qPCR. The results showed that M1 macrophages with increased ratio of iNOS/Arg1 clearly inhibited JEV infection as compared with PBS-treated macrophages. In contrast, M2 macrophages with decreased ratio of iNOS/Arg1 showed no effect on JEV infection in comparison of PBS-treated controls. These results demonstrated that macrophages with different polarized phenotypes played different roles in JEV infection. Furthermore, the up-regulation of NO in M1 macrophages mainly contributed to inhibition of JEV infection. Thus, this study provided the basis of better understanding the role of macrophages in JEV infection.
