5种鸢尾属药用植物甲醇提取物体外抗炎活性研究

【摘要】 目的研究5种鸢尾属药用植物(膜苞鸢尾、细叶鸢尾、蓝花喜盐鸢尾、粗根鸢尾、中亚鸢尾)根及根状茎甲醇提取物的体外抗炎活性。方法制备5种鸢尾属植物根及根状茎的甲醇提取物,并测定其总黄酮含量。采用脂多糖(LPS)刺激巨噬细胞RAW 264.7建立细胞炎症模型,观察5种鸢尾属植物提取物对NO、TNF-α、IL-6水平的影响。使用MTT测定细胞活力,Griess法测定NO的含量,ELISA法测定TNF-α及IL-6的含量。结果与模型组相比,5种提取物中,膜苞鸢尾及中亚鸢尾在12.5~100μg·mL-1,粗根鸢尾及蓝花喜盐鸢尾在25~100μg·mL-1,细叶鸢尾在50~100μg·mL-1时,均能有效抑制巨噬细胞释放NO(P <0.05)。膜苞鸢尾、粗根鸢尾、中亚鸢尾、蓝花喜盐鸢尾在25~100μg·mL-1,细叶鸢尾在50~100μg·mL-1时,均能有效抑制巨噬细胞释放TNF-α(P <0.05)。5种鸢尾在25~100μg·mL-1时,均能有效抑制巨噬细胞释放IL-6(P <0.01)。组间对比表明,膜苞鸢尾在100μg·mL-1时对NO和TNF-α的抑制作用最强,细叶鸢尾在25μg·mL-1时对NO的抑制作用最弱,在100μg·mL-1时对IL-6的抑制作用最弱(P <0.05)。结论 5种鸢尾属药用植物根及根状茎的甲醇提取物具有潜在的抗炎活性,本研究结果可为该属药用植物抗炎活性的研究提供参考。

【Abstract】 Objective To determine the in vitro anti-inflammation effect of methanol extracts from the roots and rhizome of 5 Iris medical plants(Iris scariosa Willd. ex Link., Iris halophila Pall. var. sogdiana(Bge.) Grubov, Iris tenuifolia Pall., Iris tigridia Bunge ex Ledeb., and Iris bloudowii Ledeb.). Methods Methanol extracts from the roots and rhizome of 5 species of Iris were prepared, and the total flavonoid contents were determined. LPSinduced RAW 264.7 macrophage was used as an experimental inflammatory model to observe the influence of 5 Iris plants extracts on the level of nitric oxide(NO), tumor necrosis factor α(TNF-α) and interleukin-6(IL-6). The proliferation of the cells was assessed by an MTT assay, and NO production was determined by Griess method. The levels of TNF-α and IL-6 were determined with sandwich ELISA. Results The extracts of Iris scariosa Willd. ex Link. and Iris bloudowii Ledeb. at 12.5-100 μg·mL-1, and Iris tigridia Bunge ex Ledeb. and Iris halophila Pall. var. sogdiana at 25-100 μg·mL-1, Iris tenuifolia Pall. at 50-100 μg·mL-1 obviously inhibited the NO release by RAW264.7 cells with significant difference(P <0.05) compared with the model group. The extracts of Iris scariosa Willd. ex Link., Iris tigridia Bunge ex Ledeb., Iris bloudowii Ledeb., Iris halophila Pall. var. sogdiana at 25-100 μg·mL-1, and Iris tenuifolia at 50-100 μg·mL-1 effectively inhibited the TNF-α release by RAW 264.7 cells. The extracts from all the 5 Iris species obviously inhibited IL-6 production. Among the 5 species, Iris scariosa Willd. ex Link. showed strongest inhibition on NO and TNF-αrelease while its methol extracts at 100 μg·mL-1, Iris tenuifolia Pall. showed the weakest inhibition on NO as well as IL-6 while its methol extracts at 25 μg·m L-1 and 100 μg·mL-1 respectively(P < 0.05). Conclusion Methanol extracts from roots and rhizome of the 5 Iris species possess anti-inflammatory property, This study can provide reference for the further research on the anti-inflammatory effect of Iris medical plants.
