氨基羟乙酸对慢性酒精中毒大鼠海马杏仁核内硫比氨和胱硫醚-β-合成酶含量及学习记忆能力的影响

【摘要】 目的探究氨基羟乙酸(AOAA)对慢性酒精中毒大鼠学习记忆能力及海马组织、杏仁核组织内硫化氢(H2S)浓度和胱硫醚β-合酶(CBS)表达的影响。方法 60只SD雄性大鼠随机分为3组,除正常对照(NC)组外,慢性酒精中毒模型(M)和AOAA治疗(AR)组给予含6%(V/V)酒精的水溶液喂养28 d。14 d后,AR组腹腔注射AOAA,1次/d,连续14 d。随后测试各组学习记忆能力,检测海马和杏仁核组织H2S的浓度和CBS表达。结果与NC组相比,M组第2~4天潜伏期、第2~4天游泳距离、海马组织H2S含量、杏仁核组织H2S含量、海马组织及杏仁核组织CBS平均光密度值均显著升高(P<0.01)。与M组相比,AR组以上各指标均显著降低(P<0.01)。结论 AOAA能够减轻酒精对大鼠学习记忆能力的影响,可能与AOAA影响海马、杏仁核内H2S浓度和CBS的表达有关。

【Abstract】 Objective To study the effect of aminooxyacetic acid(AOAA) on learning and memory as well as the content of hydrogen sulfide(H2 S) and expression of cystathionine β-synthase(CBS) in the amygdala and hippocampus in rats with chronic alcoholism. Methods 60 SD male rats were randomly divided into 3 groups,except the control group, all other groups were fed 6%(v/v) alcohol solution as the only beverage for 28 days.After 14 days, the AOAA treatment group was given medicine once a day for 14 consecutive days.The learning and memory abilities were then tested, as well as the contents of H2 S and CBS in hippocampal and amygdala tissues.Results Compared with those of blank control group, incubation period from the 2 nd to the 4 th day, swimming distance from the 2 nd to the 4 th day, the hippocampal H2 S content, the amygdala H2 S content,the hippocampus CBS average optical density value and the amygdala CBS average optical density value in the positive expression were all increased in model group, there were significant differences(P<0.01). Compared with model group, the incubation period from the 2 nd to the 4 th day, swimming distance from the 2 nd to the 4 th day, the content of H2 S in hippocampus, the content of H2 S in amygdala, the mean optical density of CBS in hippocampus, the mean optical density of CBS in amygdala were all decreased in AOAA treatment group, there were significant differences(P<0.01). Conclusions AOAA could reduce the effect of alcohol on learning and memory in rats,which might be related to the influence of AOAA on H2 S content and the expression of CBS in hippocampus and amygdala.
