高密度脂蛋白模拟肽修复肿瘤坏死因子α诱导的内皮祖细胞损伤
High density lipoprotein mimic peptide repairs the damage of endothelial progenitor cells functions induced by tumor necrosis factor α
【摘要】 目的观察高密度脂蛋白模拟肽(Rev-D4F)对肿瘤坏死因子(TNF)-α诱导的内皮祖细胞凋亡和黏附的影响。方法差速贴壁法培养内皮祖细胞;吸附低度脂蛋白和结合凝集素方法鉴定内皮祖细胞;分别给予Rev-D4F(50μg/ml)和TNF-α(30 ng/ml)进行处理,采用黏附测定实验和流式检测内皮祖细胞的黏附功能和凋亡情况。结果 TNF-α促进内皮祖细胞凋亡,但TNF-α对内皮祖细胞黏附并没有作用,Rev-D4F预先干预能明显抑制TNF-α诱导的内皮祖细胞凋亡,提高内皮祖细胞黏附功能。结论 Rev-D4F抑制TNF-α诱导的内皮祖细胞凋亡,提高内皮祖细胞黏附功能。
【Abstract】 Objective To study the effects of high density lipoprotein mimic peptide Rev-D4 F on the apoptosis and adhesion of endothelial progenitor cells(EPCs) induced by tumor necrosis factor(TNF)-α.Methods EPCs were cultured by differential time attachment and identified by adsorption of low lipoprotein and binding lectin. EPCs were treated with Rev-D4 F(50 μg/ml) and TNF-α(30 ng/ml) respectively, the adhesion function and apoptosis of EPCs were measured by adhesion assay and flow cytometry.Results TNF-α promoted EPCs apoptosis, but had no effect on EPCs adhesion. Rev-D4 F pretreatment significantly inhibited TNF-α induced EPCs apoptosis and improved endothelial progenitor cell adhesion function.Conclusions Rev-D4 F could inhibit the apoptosis of EPCs induced by TNF-α and improve the adhesion function of EPCs.
