蒙药乳腺-Ⅰ号对大鼠乳腺增生组织中膜联蛋白 A1、A2和翻译调节肿瘤蛋白表达的影响
Effect of M-I on the expression of ANXA1, ANXA2 and TCTP proteins in hyperplasia of rat mammary gland
【摘要】 目的观察蒙药乳腺(M-I)号对乳腺增生大鼠乳腺组织中膜联蛋白(ANX)A1、ANX A2和翻译调节肿瘤蛋白(TCTP)的影响。方法雌性未孕Wistar大鼠(180~220 g)36只,随机选出24只作为模型组,连续25 d肌肉注射苯甲酸雌二醇[0.5 mg/(kg·d)],再连续5 d肌肉注射黄体酮[4 mg/(kg·d)],其余8只作为正常组肌肉注射橄榄油。模型大鼠随机分为5组:模型组,给予生理盐水灌胃;阳性药对照组,给予三苯氧胺1.8 mg/kg灌胃;M-I号低剂量组和M-I号高剂量组别给予M-I号0.5 g/kg和3.0 g/kg灌胃。给药结束后,观察大鼠乳腺组织病理学特征,检测大鼠血清及乳腺组织中ANXA1、ANX A2、TCTP水平。结果与正常组大鼠比较,模型组大鼠乳腺组织中ANX A1、ANX A2、TCTP表达水平均显著升高(P<0.05),乳腺组织增生性改变明显。与模型组大鼠比较,M-I号高剂量组乳腺组织中ANX A1、ANX A2、TCTP表达水平显著降低(P<0.05),乳腺组织增生性改变较模型组明显减轻。结论 M-I号对乳腺增生大鼠有较好的治疗作用,其治疗机制涉及对ANX A1、ANX A2、TCTP的调节作用。
【Abstract】 Objective To observe the effects of Mongolian medicine mammary-I(M-I) on Annexin A1(ANXA1), Annexin A2(ANXA2)and translationally controlled tumor protein(TCTP)in the breast tissue of breast hyperplasia rats. Methods From 36 unfertilized female Wistar rats, 8 were randomly selected as normal control group, the rest of the animals were treated with muscle injection of estradiol benzoate(0.5 mg/kg) everyday for 25 days, then were treated with muscle injection of progesterone(4 mg/kg) everyday for 5 days to copy of mammary gland hyperplasia. Rasts in normal control group were treated with muscle injection of saline solution. After successful model replication, model rats were randomly divided into model, positive control(1.8 mg/kg tamoxifen was administered to the stomach), low-, high-dose M-I(0.5 and 3.0 g/kg M-I were given by gavage) groups. After the administration, the pathological features of the breast tissue of rats were observed, and ANXA1, ANXA2 and TCTP in the serum and breast tissue of rats were detected. Results Compared with normal control group, ANXA1, ANXA2 and TCTP expression in breast tissue of the model group were significantly increased(P<0.05), and hyperplasia of breast tissue was significantly changed. Compared with model group, ANXA1, ANXA2 and TCTP expression in breast tissue of rats in the high-dose M-I group were significantly reduced(P<0.05).Conclusions M-I has a good therapeutic effect on hyperplasia rats, and its therapeutic mechanism involves the regulation of ANXA1, ANXA2 and TCTP.
