EGCG对人肝星状细胞的生长状态及活化表型的影响
Effects of EGCG on proliferation and activation of human hepatic stellate cells
【摘要】 目的研究表没食子儿茶素没食子酸酯(EGCG)对体外培养的人肝星状细胞株(LX-2)生长状态及活化表型的影响。方法利用实时无标记细胞分析技术系统连续监测72h不同浓度EGCG(0、25、50、75、100、125、150、175μmol/L)对LX-2细胞生长状态的影响;通过流式细胞仪检测LX-2细胞的凋亡率和活化标志物α-平滑肌激动蛋白(α-SMA)的表达变化;采用吖啶橙/溴乙锭染色法在倒置荧光显微镜下观察EGCG处理后的LX-2细胞形态改变。结果随药物浓度增加,EGCG对LX-2细胞的增殖抑制作用增强,细胞增殖速率降低(P<0.05);EGCG能诱导LX-2细胞的凋亡(P<0.05),同时抑制α-SMA的表达(P<0.01)。结论EGCG能通过诱导细胞凋亡的方式抑制LX-2的增殖和活化。
【Abstract】 Objective To investigate the effects of epigallocatechin-3-gallate(EGCG) on the proliferation and activation of human hepatic stellate cells in vitro. Methods Real-time unlabeled cell analysis(RTCA) system was used to continuously monitor the growth status of human hepatic stellate LX-2 cells treated with different concentrations of EGCG(0, 25, 50, 75, 100, 125, 150, 175μmol/L) for 72 h.The apoptosis and the expression of activation marker-smooth muscle activin(α-SMA) in LX-2 cells were detected by flow cytometry. The morphological changes of LX-2 cells after EGCG treatment were observed under fluorescence microscope by AO/EB staining. Results With the increase of drug concentration, EGCG significantly inhibited the growth of LX-2 cells and decreased the proliferation rate(P<0.05).EGCG significantly induced the cells apoptosis and suppressed α-SMA expression(P<0.01). Conclusion EGCG can inhibit the proliferation and activation of LX-2 cells by inducing apoptosis.
