胆碱膳食应激小鼠肠道菌群及代谢
Gut microbiota and metabolism in mice with choline dietary stress
【摘要】 研究质量分数为1%的高胆碱膳食对健康小鼠肠道菌群组成及代谢的影响。选取20只C57BL/6J小鼠,随机分成正常饮食组(CK)和1%高胆碱膳食组(CHO)。处理60 d后,采用宏基因组测序技术和气相色谱法(Gas chromatography,GC)分别测定小鼠粪便肠道菌群组成和短链脂肪酸(Short chain fatty acids,SCFA)的质量分数;ELISA试剂盒测定小鼠血浆脂多糖(Lipopolysaccharides,LPS)浓度;原子吸收分光光度法测定粪便微量元素质量分数。结果显示:高胆碱饮食改变肠道菌群组成,显著增加毛螺菌属NK4A136组和氧化三甲胺(Trimethylamine oxide, TMAO)产生菌梭菌目的丰度;与CK组相比,CHO组粪便中乙酸质量分数显著增加,而血浆LPS浓度明显降低,同时,1%胆碱饮食处理后粪便中Fe的质量分数显著提高。因此对于健康小鼠,摄入1%胆碱膳食能改变肠道菌群组成,提高TMAO相关菌丰度。此外,1%胆碱膳食可提高乙酸含量,降低LPS的水平,具有保护肠黏膜屏障的功能。
【Abstract】 In this study, the influences of 1% choline dietary on gut microbiota composition and metabolism in health mice were investigated. 20 mice(C57BL/6J) were randomly divided into two groups: normal diet group(CK) and 1% choline diet group(CHO). After 60 d, metagenomic sequencing, gas chromatography(GC) and atomic absorption spectrophotometry were used to analyze the composition of gut microbiota, the content of short chain fatty acids and the content of trace elements in the feces samples of the mice, respectively. ELISA kit was utilized to determine the content of lipopolysaccharide in plasma lipids of the mice. The results showed that high choline diet could change the composition of intestinal microflora and greatly increase the ratios of Lachnospiraceae NK4A136 group and bacteria-Clostridiales produced from trimethylamine-oxide(TMAO). The content of acetic acid and Fe in feces of CHO group were significantly higher than that of CK group. Meanwhile, 1% choline treatment could significantly reduce the levels of LPS in plasma. Therefore, for healthy mice, consumption of 1% choline diet could improve the composition of gut flora, increase the abundance of bacteria produced from TMAO, increase the concentration of acetic acid and reduce the LPS level, thereby protecting their intestinal mucosal barrier.
