贵州禾来拢茎尖遗传转化体系的研究

【摘要】 以贵州禾来拢茎尖为材料,遗传转化含有Ubiqutin启动子的苦瓜几丁质酶基因McCHIT1,设计正交试验,探究农杆菌菌液浓度、真空渗透压和真空处理时间对贵州禾来拢茎尖遗传转化的影响。结果表明:对植株的死亡率及转化率影响最大的是真空处理时间,其次为菌液浓度,真空渗透压的影响较小。综合考虑植株死亡率及转化效率认为,菌液侵染浓度OD600 0.8,真空处理时间11 min,真空渗透压16 kPa为农杆菌介导贵州禾来拢茎尖遗传转化的最优参数。

【Abstract】 In the study,we used the Guizhou He Lailong’s shoot tips as the research material to transform McCHIT1 containing the Ubiqutin promoter,and investigated the influence of the concentration of agrobacterium culture solution,vacuum osmotic pressure and vacuum treatment time on the efficiency of agrobacterium-mediated genetic transformation of Guizhou He Lailong’s shoot tips by orthogonal experimental design method.The results showed that the vacuum treatment time had the greatest influence on plant mortality and conversion rate,followed by the concentration of bacteria solution,and the vacuum osmotic pressure had little influence.According to the comprehensive consideration of plant mortality and transformation efficiency,the optimal parameters for agrobacteria-mediated genetic transformation of the shoot tip of Guizhou He Lailong were the infection concentration OD600 0.8,vacuum treatment time 11 min and vacuum osmotic pressure 16 kPa.
